冠突散囊菌菌落计数方法的优化

         

摘要

茯砖茶中冠突散囊菌(Eurotiumcristatum)的数量是评判其产品质量的重要因素,目前大多数检测机构均使用传统的国标法来检测冠突散囊菌的菌落数量.冠突散囊菌有闭囊壳构造,传统国标法中计数方法存在缺陷,不能检测出茯砖茶中冠突散囊菌的真实数量.通过对传统方法的优化和改进,对样品充分粉碎并过60目筛,利用200颗玻璃珠增加振摇强度,延长振摇时间至60min,从而打破冠突散囊菌闭囊壳,释放其中的子囊孢子,能获得茯砖茶中所含冠突散囊菌的真实数量.结果表明:该方法优化效果明显,操作简单、经济实用性好.%The number of Eurotiumcristatum in Fuzhuan tea is an important factor in judging the quality of tea products. The majority of the testing institutions used the traditional national standard method to detect the number of Eurotiumcristatum. However, it contains cleistothecium and the traditional national standard method could under-estimate the real quantity of the microbes. There were several ways applied to improve the traditional method , such as increasing the crush of raw materials, improving strength of shaking process with 200 glass beads and prolonging the shaking time to 60 minutes. It can break cleistothecium and release ascospores. The method of plate culture counting was optimized so as to obtain the true quantity of the Eurotiumcristatum that contained in Fuzhuantea . The results showed that optimized method reached obvious effects with simply and economically.
