酶法辅助提取牦牛血中超氧化物歧化酶的r工艺条件优化

         

摘要

The superoxide dismutase (SOD) was extracted by single factor test and response surface test to opti-mize the enzymatic extraction of SOD from yak blood cells. With ultrasonic and pancreatin co-action of blood cells for hemolysis treatment, the specific activity of SOD as an indicator to test, The results showed that the optimum conditions were as follows: enzymolysis temperature 40℃, enzymolysis time 110 min, enzymolysis pH 7.5, enzyme addition 0.6% under ultrasonic power fixed 300W. The activity of SOD was 1410.62U/mg. Under this process, fur-ther extraction of SOD could significantly increase the specific activity of SOD.%以牦牛血血细胞为原料,通过单因素试验和响应面试验优化酶法提取超氧化物歧化酶(SOD).用超声波和胰酶共同作用血细胞进行溶血处理,以SOD的比活力为指标进行试验,结果表明在超声功率固定300W条件下,得到最优工艺条件为:酶解温度40℃、酶解时间110min、酶解pH7.5、酶添加量0.6%.所得SOD比活力为1410.62U/mg.在此工艺条件下,提取SOD可显著提高SOD比活力.
