目的:探讨乌司他丁对缺血再灌注脑损伤的保护作用。方法:选取健康雄性SD大鼠72只,将所有大鼠以随机数字表法分为假手术组、缺血再灌注损伤模型组和乌司他丁处理组,每组各24只。建立大鼠缺血再灌注模型,检测3组大鼠脑组织紧密连接蛋白1(zonula occluden-1,ZO-1)及血清神经元特异性烯醇化酶(NSE)的表达变化。结果:与缺血再灌注损伤模型组大鼠相比,乌司他丁处理组大鼠脑组织中ZO-1蛋白表达显著增加,NSE表达明显减少,差异均有统计学意义(P<0.05)。结论:乌司他丁可通过抑制脑缺血再灌注后血-脑脊液屏障通透性的增加、减少NSE表达而发挥脑保护作用,其保护血-脑脊液屏障的机制可能与增加ZO-1蛋白的表达有关。%OBJECTIVE:To probe into the protective effects of ulinastatin on ischemia-reperfusion injury. METHODS:72 healthy male SD rats were randomly selected to be divided into sham operation group, schemia-reperfusion injury model group and ulinastatin group, with 24 cases in each.Schemia-reperfusion injury model was established, expression changes of zonula occluden-1(ZO-1) and neurone specific enolase(NSE) in brain tissues of three groups were detected.RESULTS: Compared with schemia-reperfusion injury model group, expression of ZO-1 protein in ulinastatin group significantly increased, while NSE decreased significantly, with statistically significant difference(P<0.05).CONCLUSIONS:Ulinastatin can play a protective role in the brainm by inhibiting the increased cerebral ischemia and reperfusion brain barrier permeability, and reducing expression of NSE;and its protective blood-brain barrier may be related to upregulation expression of ZO-1 protein.
