探索了提高枯草芽孢杆菌产生芽孢的方法,使用单批一次投料培养法研究了培养基成分、pH值、培养过程中通气量及装液体积比对菌体产芽孢的影响.试验结果说明:枯草芽孢杆菌最适生长和产生芽孢的碳源是糖蜜,最适氮源是酵母浸粉;二价锰离子和无机盐对菌体的生长和产芽孢有很大影响.菌体生长和产芽孢的最适培养基组成:糖蜜1.0%,酵母粉0.8%,(K2HPO4+KH2PO4) 0.5‰,MnSO40.2‰;产芽孢的最适条件:开始培养pH值为7.0,最适装液量为20%(体积比).在5L半自动发酵罐中,溶氧大于70%,发酵24 h后,芽孢数量为5.1×1011CFU/mL,出芽率为98.0%.%The method to improve the Bacillus subtilis producing spores was investigated.Effect of culture medium composition,pH value,ventilation during culture and liquid volume ratio on the Bacillus subtilis producing spores was studied by using a single batch feeding culture method. The results showed that suitable carbon source and nitrogen source for Bacillus subtilis growth and producing spores were molasses and yeast extract.The divalent manganese ions and inorganic salts had a great influence on the bacteria growth and producing spores. The optimal medium of bacterial growth and spore production was as follows: 1.0% molasses, 0.8% yeast powder, 0.5‰ K2HPO4and KH2PO4, 0.2‰MnSO4; Optimal conditions for producing spores were as follows: in the initial training, the pH 7.0 and the optimal loading capacity 20% (volume ratio). In the 5 L semi-automatic fermentation tank, dissolved oxygen was more than 70%, and after fermentation, the number of spores was 5.1×1011CFU/mL, and the budding rate was 98.0%.
