楤木皂苷通过降低 Akt/NF-κB 通路抑制HeLa 细胞的增殖并促进其凋亡

             

摘要

Objective To explore the effect of araloside on the proliferation and apoptosis of HeLa cells.Methods Human cervical cancer cell line HeLa was cultured in vitro,the experiment was divided into 4 groups as follows:blank group,araloside trea-ted groups(50,100,200 μg/mL).Normal saline and araloside (50,100,200 μg/mL)were gave,respectively.24,48 and 72 hours lat-er,the cells were collected.Cell proliferation was detected by MTT,cell apoptosis was determined by flow cytometry,the expression of pAkt1,pIкBα,NF-κB (p65),Bcl-2 and Caspase-3 were evaluated by western blot.Results Araloside obviously inhibited the pro-liferation and increased the apoptosis level of HeLa cells in a time-dose dependent manner.Moreover,Araloside significantly de-creased the phosphorylation of Akt1 and IκBα,reduced the expression of NF-κB(p65)and Bcl-2,whereas obviously increased Caspase-3 content.Conclusion Araloside could inhibit the proliferation and promote the apoptosis of HeLa cells via suppressing Akt/NF-кB signaling pathway.%目的:探讨!木皂苷对人宫颈癌 HeLa 细胞增殖和凋亡的影响。方法以体外培养的人宫颈癌 HeLa 细胞为研究对象,实验分为阴性对照组和!木皂苷组(50、100、200μg/mL)共4组,分别给予 RPMI-1640培养液和!木皂苷(50、100、200μg/mL)处理。24、48和72 h 后,分别收集各组细胞,采用 MTT 法检测细胞的增殖情况,流式细胞仪检测细胞凋亡,Western blot 技术测定细胞内蛋白(pAkt1、pIкBα、NF-κB p65、Bcl-2和 Caspase-3)表达水平。结果!木皂苷呈时间-剂量依赖性地抑制 HeLa 细胞的增殖,并增加其凋亡水平。另外,!木皂苷显著降低 Akt1和 IкBα的磷酸化水平,减少 NF-κB(亚基 p65)和 Bcl-2的蛋白表达,相反却显著增加 Caspase-3的含量。结论!木皂苷通过降低 Akt/NF-κB 信号通路的活性抑制宫颈癌 HeLa 细胞的增殖并促进其凋亡。
