组织因子途径抑制物基因多态性在肺栓塞中的研究

     

摘要

目的 探讨组织因子途径抑制物(TFPI)基因多态性与肺血栓栓塞症(PTE)的关系.方法 选择PTE患者35例,健康对照组34名.采用聚合酶链反应(PCR/RT-PCR)检测PTE组和健康对照组TFPI C-399T、T-287C基因型及等位基因频率,结果经测序证实.结果 PTE组C-399T纯合突变型TT、杂合子突变型CT和正常纯合子CC与健康对照组之间差异无统计学意义(P>0.05);T和C等位基因型频率与健康对照组比较差异无统计学意义.PTE组T287-C杂合子突变型CT、纯合突变CC和正常纯合子CC与健康对照组比较差异无统计学意义(P>0.05);T和C等位基因频率分布与对照组比较,差异也无统计学意义(P>0.05).结论 TFPI基因C-399T、T287-C多态性可能与PTE无相关性.%Objective To explore the correlation between the gene polymorphism of tissue factor pathway inhibitor (TFPI) and pulmonary thromboemlism (PTE). Methods Thirty-five PTE patients were recuited, with 34 healthy people assigned as normal controls. TFPI C-399T and T-287C genotypes and the allele frequencies were detected in the PTE group and the normal control group by RT-PCR. The results were then verified by sequencing. Results There was no significant difference in C-399T homozygous mutant TT, heterozygote-mutant CT and normal homozygote CC between PTE and normal control groups (P>0.05). No statistical difference was found in T and C allele frequencies between these two groups. Compared with the normal control group, PTE group showed no statistaical difference in T287-C heterozygote-mutant CT, homozygous mutant CC and normal homozygote CC (P>0.05). In addition, no difference was found in distribution of T and C allele frequencies in the two groups (P>0.05). Conclusion There might be no correlation between gene polymorphism of TFPI genetypes (C-399T plus T287-C) and PTE.
