铁包金总黄酮体内对S180实体瘤的抑制作用

         

摘要

目的 研究铁包金总黄酮对小鼠移植性肿瘤S180的生长抑制作用,并探讨其可能的作用机制.方法 用动物移植性肿瘤在体实验法,观察抑瘤率、胸腺脾脏及肝脏指数;检测血清中SOD、MDA变化;病理切片观察瘤细胞生长及病理形态变化情况;免疫组化检验肿瘤组织中p53、TNF-α、Caspase-3蛋白的表达.结果 ① 铁包金总黄酮高、中、低剂量组的抑瘤率分别50.35%、35.66%、23.08%,阳性对照组的抑瘤率为83.22%,各组小鼠的胸腺、脾脏、肝脏指数与空白组比较差异无显著性(P>0.05),阳性对照组与空白组比较明显降低(P<0.01);② 铁包金总黄酮各组升高小鼠血清的SOD值,且降低其MDA值,具有一定的量效关系.阳性组降低小鼠的SOD值,且升高MDA值.与空白组相比差异有显著性(P<0.01);③ 病理切片显示给药各组和阳性组坏死面积比空白组大;④ 免疫组化显示高、中、低各组p53蛋白表达依次升高,TNF-α和Caspase-3蛋白表达依次降低.结论 铁包金总黄酮通过清除氧自由基和调节p53,TNF-α和Caspase-3蛋白的表达来抑制肿瘤生长.%Aim To study the anti-tumor effects of the flavone from Berchemia lineata( L )DC in vivo and discuss its possible mechanisms.Methods A total of 50 cases of S180 mice were assigned randomly into 5 groups : saline group( control ), Cisplatin group( positive control )and flavone high, middle and low group.The anti-tumor effect on internal organ of mice was observed to study the tumor inhibition rate in different groups.Influence of flavone on mouse immune system was studied by comparing the index of thymus, spleen and liver.SOD & MDA was detected in serum.The growth and patho-morphologic changes of tumor cells were observed through pathological sections.The protein expression level of p53, TNF-a and Caspase-3 were detected by immunohistochemistry.Results ①The inhibitory rate was 50.35%,35.66% ,23.08% in the flavone group and 83.22% in the positive control group respectively.Mouse thymus , spleen , liver index in flavone group had no significant difference compared with that of the control group( P> 0.05 ).However, positive thymus spleen,liver index in control group had a significant decrease compared with that in other groups ( P< 0.01 ).② The flavone group of mouse SOD significantly increased and MDA reduced.③ Pa thology revealed that necrotic area of dose groups and the positive group was larger than the control group.④ Immunohistochemistry showed that p53 protein expres sion was increased in dose groups , TNF-α and Caspase 3 protein expression decreased gradually.Conclusion The flavone can scanvenge oxygen free radicals and adjust express of p53, TNF-α and Caspase-3 to sup press tumor growth.
