猪狂犬病的确证

     

摘要

To confirm the clinically suspected case of pinges infected with rabies virus in a small hoggery in Guangdong Province, the brain tissue samples including cerebrum, cerebellum and hippocampal gyrus were subjected to Giemsa stain,fluo rescent antibody test (FAT) and RT PCR. The samples were inoculated to suckling and adult mice. The results indicated that the inclusion bodies and a lot of green focus could be found in hippocampal gyrus, and a 424bp DNA fragment of rabies virus N gene could be detected from the brain tissues. Between 13 and 15 days post infection, clinical syptoms of a rabies like illness developed in all inoculated mice. It was suggested that the pigs in the small hoggery were infected with rabies virus.%目的 了解广东省-猪场发生狂犬病猪只的感染状况.方法 采集疑似狂犬病发病猪的大脑、小脑以及海马回,制成切片,用姬姆沙染色,在光学显微镜下检测狂犬病病毒包涵体.用抗狂犬病病毒N蛋白荧光抗体染色,在荧光显徽镜下检测特异荧光.用蛋白酶K处理病料,抽提核酸,以狂犬病病毒N基因特异引物作RT-PCR检测狂犬病病毒特异性核酸.将病料接种小白鼠接毒,观察小鼠的发病.结果 病理学切片检测病猪脑海马神经细胞中有包涵体;脑组织印片经特异性抗狂犬病病毒核蛋白(NP)单克隆抗体免疫荧光染色后,荧光显微镜下可见大量的针尖状绿色荧光颗粒;狂犬病病毒NP特异性目的 基因RT-PCR得到与预期结果大小一致的扩增产物;小白鼠接毒试验,3日龄以内乳鼠接毒后出现特异性的神经症状.结论该猪场病死猪检测狂犬病病毒感染阳性,确定猪群中发生狂犬病.
