微波辐射致大鼠海马损伤中MAPK传导通路的差别激活

     

摘要

AIM: To discuss the effects of microwave irradiation on the neurocyte mitrogen-activated protein kinases(MAPK) signal conductive system in hippocampus nerve and the reaction of MAPK conductive gateway in hippocampus neural damages.METHODS: Anti-phosphorylation extracellular synal-regualted kinase (ERK1/2), C-Jun N-terminal kinase(JNK) and P38 MAPK antibody were introduced in Western blot process to dynamically observe the phosphorylation changes of rat hippocampus neurocyte protein ERK, JNK/SAPK and P38 MAPK in order to reflect the functional status of MAPK signal gateway after microwave irradiation.RESULTS: Within 12 hours after microwave irradiation, ERK and JNK were continuously activated, the peak of ERK appeared immediately after irradiation, and JNK peak appeared after 3 hours of irradiation. And after 24 hours,both activations were inhibited; P38 MAPK was continuously in activating status within 24 hours after irradiation, and there were 2 activating peak appeared at 3 and 24 hours after irradiation.CONCLUSION: Microwave irradiation can induce differentiate activation in three MAPK signal conductive gateways of neurocyte of hippocampus and moreover, the hippocampus damages induced by microwave irradiation might be achieved through the differentiate activation of MAPK signal conductive gateway.%目的:探讨微波辐射对海马神经神经细胞有丝分裂素激活蛋白激酶类(mitogen-activated protein kinases,MAPK)信号转导系统的影响,以及MAPK信号转导通路在微波辐射导致海马神经损伤中的作用.方法:采用抗磷酸化形式的细胞外调节蛋白激酶(extracellular synal-regulated kinase,ERK1/2),C-Jun氨基末端激酶(C-Jun N-terminal kinase,JNK),P38 MAPK抗体,进行Western blot,动态观察微波辐照后大鼠海马神经细胞ERK,JNK/SAPK,P38MAPK蛋白质磷酸化情况,以此反映MAPK信号通路的功能状态.结果:微波辐照后12 h内ERK,JNK持续激活,ERK的激活高峰在辐照后即刻,JNK激活高峰出现于辐照后3,24 h后两者活化均受抑制;P38MAPK辐射后24 h内一直处于激活,并在3,24 h出现两次激活高峰.结论:微波辐射可使海马神经细胞3条MAPK信号转导通路出现差别激活,并且微波辐射导致的海马神经损伤可能是通过MAPK信号转导通路的差别激活实现的.
