鸭疫里默氏杆菌假定溶血素相关蛋白基因Riean _0620的原核表达及溶血活性检测

摘要

To investigate the biological function of hypothetical hemolysin-related protein Riean_0620 of R.anatipestifer,Riean_0620 gene from serotype 10 R.anatipestifer strain HXb2 was amplified by PCR and sequenced.Then,the gene ORF was inserted into the prokaryotic expression vector pET-43,la.The expression of recombinant protein Riean_0620 was induced with IPTG and rRiean-0620 was purified with affinity chromatograph.The hemolytic activity assay showed the recombinant protein rRiean_0620 had the ability to lyse duck erythrocytes.The result of this study benefit to further study on biological functions of Riean 0620 protein and hemolysis mechanism of R.anatipestifer.%为研究鸭疫里默氏杆菌(RA)的假定溶血素相关蛋白Riean 0620的功能,本研究根据血清10型RAHXb2株Riean 0620基因序列设计引物,PCR扩增Riean 0620基因,并进行序列测定与分析.将Riean 0620基因克隆到原核表达载体pET-43.1a进行诱导表达,采用亲和层析纯化获得重组蛋白rRiean 0620.溶血活性检测结果表明rRiean 0620具有裂解鸭红细胞的活性.本研究将有利于进一步研究Riean 0620蛋白的生物学特性以及RA的溶血机制.
