大豆膨胀素GmEXPA4基因的克隆及表达分析

     

摘要

Degenerate primers were designed according to expansin gene sequences from other plants. Conserved sequence of expansin gene in Glycine max L. was cloned using young shoot cDNA of a dwarf mutant HK808. Full-length expansin gene, designated as GmEXPA4, was cloned by RACE. Bioinformatics result showed that GmEXPA4 was a member of alpha-expansin subfamily. Fluorescence real-time quantification PCR indicated that GmEXPA4 expression was constitutively in different period in wildtype Dongnong 42 and mutant HK808. The relative expression showed close relation to the amount of soybean growth.%根据植物膨胀素基因保守区设计简并引物,以大豆矮化突变体HK808幼芽的cDNA为模板,克隆到530bp的膨胀素基因保守区片段,并结合RACE技术克隆得到膨胀素基因全长序列,将其命名为GmEXPA4基因.生物信息学分析结果表明,该基因属于膨胀素中的α- expansin亚家族.GmEXPA4基因的实时荧光定量相对表达分析表明,该基因在东农42及其矮化突变体HK808中的各个时期都有表达,且与植物的生长量密切相关.
