高效液相色谱-荧光检测法测定人血浆中洛美沙星的浓度

     

摘要

Objective To apply high performance liquid chromatography-fluorescence assay (HPLC-FLU) in measuring concentration of lomefloxacin in human plasma. Methods Plasma samples were precipitated by methanol. Methanol-monopotassium phosphate buffer solution (pH: 2.8, 35:65, v/v) was applied as mobile phase, and its flow velocity was 1.0 ml/min, chromatographic column was BaiAn-C18(4.6×150 mm, 5 μm), column temperature was 25℃, and Fluorescence detection wavelength was λex=280nm and λem=455nm. Results The intrinsic impurity of plasma did not interfere with the measurement of determinand. Linearity of lomefloxacin was good at 0.10~10.04μg/ml (r=0.9995), and the lower limit of quantitation was 0.10 µg/ml. The day-to-day and within-day relative standard deviation (RSD) were all less than 10%. Conclusion This measure method is fast, sensitive, and simple, which can be applied to the pharmacokinetic study of lomefloxacin.%目的:建立用高效液相色谱-荧光检测法(HPLC-FLU)测定人血浆中洛美沙星的浓度。方法血浆样品经甲醇沉淀后,以甲醇-磷酸二氢钾缓冲液(pH:2.8,35:65, v/v)为流动相,流速为1.0 ml/min,色谱柱为BaiAn-C18(4.6×150 mm,5μm),柱温为25℃,荧光检测波长λex=280 nm,λem=455 nm。结果血浆内源性杂质不干扰待测物测定,洛美沙星在0.10~10.04μg/ml有良好线性关系(r=0.9995),定量下限为0.10μg/ml,日内、日间精密度(RSD)均小于10%,样品稳定性良好。结论该法灵敏、快速、简便,适合用于洛美沙星的药动学研究。
