HPLC-ELSD测定三两半药酒中黄芪甲苷的含量

摘要

目的:采用HPLC-ELSD测定三两半药酒中黄芪甲苷的含量.方法:采用Diamonsil C18色谱柱,以乙腈-水(28:72)为流动相,流速1.0ml·min-1,柱温30℃,漂移管温度45℃,载气为N2,流速3.5 ml·min-1.采用外标两点法计算含量.结果:黄芪甲苷在(0.56～11.20)mg·L-1浓度范围内有良好线性关系(r=0.9999),平均加样回收率为98.4%,RSD为0.95%.结论:本方法快速简便,结果准确,可用于该制剂中黄芪甲苷的定景分析.%Objective: To establish an HPLC-ELSD method for the determination of astragaloside Ⅳ in Sanliangban medicinal liquor. Methods: The Diamonsil C18 column was used, the mobile phase consisted of acetonitrile- water (28 : 72 .v/v), the flow rate was 1.0 mL-min-1, the column temperature was 30℃, the temperature of drift tube was 45℃, nitrogen gas as carrier gas, the flow rate was 3.5 mL-min-1, with a universal detector Evaporation Light Scattering Detector (ELSD). Calculate contents with extemal standard two-point method.Results:The linear response range of astragaloside Ⅳ was (0.56～11.20)mg·L-1 (r=0.9999). The average recovery was 98.4％, RSD was 0.95％. Conclusion: The method is sample, repeatable and accurate. it can be applied in quantitative determination of astragaloside Ⅳ in Sanliangban medicinal liquor.
