γ-氨基丁酸能神经元条件性敲除PGC-1α基因小鼠的制备

摘要

Objective To generate mice which are specific for peroxisomproliferator-activated receptor-γ coactivator-1 (PGC-1α) knockout in the GABAergic interneuron.Methods Conditional mice specific for PGC-1αf/+ were introduced from the Jackson Laboratory,USA and initially inbred to obtain homozygote PGC-1αf/f mice.The PGC-1αf/f conditional mice were further crossed with Dlx5/6-Cre-IRES-EGFP transgenic mice to achieve specific knockout of PGC-1α in the GABAergic interneuron.Results The offspring with specific knockout PGC-1α gene were successful for the generation of GABAergic interneuron,with the resulting genotype being PGC-1αf/f;Dlx5/6-Cre-IRES-EGFP.Conclusion The PGC-1αf/f;Dlx5/6-crE-IRES-EGFP mice were obtained through a proper crossing strategy,which has provided a suitable platform for studying the function of PGC-lα in neuropsychiatric diseases.%目的 制备在γ-氨基丁酸能神经元条件性敲除过氧化物酶体增殖物激活受体γ辅激活因子-1α(peroxisomproliferator-activated receptor-γ coactivator-1,PGC-1α)基因小鼠.方法 从美国Jackson Lab引进SPF级条件性敲除PGC-1α基因的杂合子小鼠PGC-1αf/+,通过饲养、繁育获得PGC-1αf/f纯合子小鼠.通过将PGC-1αf/f小鼠与在γ-氨基丁酸能神经元表达Cre重组酶的Dlx5/6-Cre-IRES-EGFP小鼠进行繁育,获得在γ-氨基丁酸能神经元条件性敲除PGC-1α的基因鼠PGC-1αf/f;Dlx5/6-Cre-IRES-EGFP.结果 成功制备了在γ-氨基丁酸能神经元条件性敲除PGC-1α的基因鼠.结论成功引进PGC-1αf/+杂合子小鼠,应用合理的基因敲除策略,获得在γ-氮基丁酸能神经元敲除PGC-1α的基因鼠,为神经精神疾病的机制研究提供了一种可信的模型动物.
