4种方法检测表皮葡萄球菌生物膜形成能力的应用价值探讨

         

摘要

目的 比较4种方法检测表皮葡萄球菌体外生物膜形成能力的应用价值,帮助临床选择检测生物膜形成能力的可靠方法.方法 用刚果红实验、半定量粘附实验、扫描电镜实验、PCR扩增icaA分别检测临床分离的45株表皮葡萄球菌生物膜的形成能力,经行×列表χ2检验比较阳性检出率,并以扫描电镜检测法为"金标准"计算其他3种方法的灵敏度和特异性.结果 4种方法检出表皮葡萄球菌生物膜形成的阳性率分别为31.1%、40.0%、40.0%、28.9%,无统计学差异(χ2=1.80,P>0.05).刚果红实验、半定量粘附实验、PCR扩增icaA检测生物膜形成能力的灵敏度分别为77.8%、100%、77.2%,而特异性均为100%.结论 4种方法均可用于体外生物膜检测.%Objective In order to choose a suitable method in detecting biofilm-fomning strains of clinical Staphylococcus epi-dermidis isolates ,the detection rate ,sensiu'vity and specificity of Congo Red agr method , semi-quantitative adherence assay, scanning electron microscopy (SEM ) and PCR for icaA gene are evaluated .Methods the biofilm formation of Staphylococcus epidermidis were detected by the four methods mentioned above in 45 clinical isolates .then comparing the postive detection rate by χ2 test and calculating sensitivity and specificity of the other three methods based on SEM as a gold standard .Results The detection rate of four methods have no difference in detecting the biofilm formation ,Sensiu'vity and of Congo Red agr method ,semrquantitative adherence assay and PCR for icaA gene in detecting biofilm-forming strains of Staphylococcus epidermidis are 77 .8% 、100% 、77 .2% respectively ,howev-er the specificity of three methods are all 100 %.Conclusion four methods can be used to detect biofilm-forming strains of Staphylo-coccus epidermidis ,whereas semi-quantitative adherence assay is relatively more suitable for biofilm detection in clinical laboratory .
