目的 探讨清开灵注射液(QKL)对心肌缺血-再灌注损伤后大鼠心肌细胞凋亡相关蛋白Caspase表达的影响及其作用机制.方法 实验共设假手术组、模型组、复方丹参注射液组(DS组)及清开灵注射液组(QKL治疗组),每组8只,各组术前连续舌下静脉给药1周,采用结扎大鼠左冠状动脉前降支40 min后,松解再灌注120 min,制备心肌缺血-再灌注模型,实验结束后HE染色观察心肌细胞形态学变化,免疫组化观察心肌Caspase-3、Caspase-9表达情况.结果 QKL能够下调心肌细胞Caspase-3、Caspase-9表达,从而减少心肌细胞的凋亡.结论 QKL 对大鼠心肌缺血-再灌注损伤具有保护作用,其保护作用的机制与下调心肌细胞凋亡相关蛋白Caspase-3、Caspase-9表达有关.%Objective To explore the effects of Qingkailing (QKL) on apoptosis in myocardia of rats after ischemic reperfusion. Methods Rats were divided into sham,model,DanShen and Qingkailing groups at random,8 rats in each, and were treated with medicines by sublingual vein administration respectively last 1 week. All rats were subjected to openning chest then the left anterior desending coronary arteries (LAD) were ligated for 40 min and undamped for 120 min,at the end of,myocardial pathological changes were observed by light microscope after HE dyeing,and measure the apoptotic cells by TUNEL to observe the expressiones of caspase 3 and caspase 9 by immunohistochemistry. Results QKL downregulated the expressiones of Caspase 3, Caspase 9. Conclusion QKL have protective effects on ischemi areperfusion in rats,which may be related to downregulated the expressiones of Caspase 3,Caspase 9 during ischemia reperfusion.
