替米沙坦对胰岛素抵抗SD大鼠内脏脂肪细胞体积的影响

         

摘要

Objective To observe the effect of telmisartan on the adipocyte size in epididymal fat tissue of Sprague Dawley(SD)rats with insulin resistance induced by a long term high sugar and fat diet(HSHFD),and to discuss the possible machanism of effect on degree of insulin resistance induced by telmisartan. Methods The male SD rats were randomly divided into three groups:the normal diet group(NC,n=10),the HSHF group (HSHF,n=6)and the HSHF+ Tel group (Tel,n=10).The rats were fed a HSHFD for 24 weeks in model groups,then the rats were intervened by telmisartan for 12 weeks in Tel group,the rats were fed as before for 12 weeks in NC group and HSHF group.Final y,in three groups,body weight (BW),fasting blood glucose(FPG),fasting serum insulin (FINS)and serum triglyceride (TG)were examined.Insulin resistance index was calculated.Meanwhile,the left epididymal adipose tissues were col ected from each rat and routinely stained using hematoxylin and eosin,the adipocyte size were observe with optical microscope.Results Compared with the NC group,BW,FPG,FINS,TG,HOMA IR in the HSHF group was significantly higher(P<0.05).Compared with the HSHF group,BW,FINS,TG,HOMA IR were significantly lower in the Tel group(P<0.05).FPG tended to be lower in Tel group,although the differences were not statistical y significant(P>0.05).The adipocyte size was almost same and smal in NC group.On the contrary,the adipocyte size was quite different and large in HSHF group.Compared with HSHF group,the adipocyte size was decreased in Tel group.Conclusion Telmisartan can prevent body weight gain and improve serum lipid metabo-lism and insulin resistance in rats fed with a long term HSHFD.Telmisartan can reduce the adipocyte size and fat content in epididy-mal fat tissue of rats fed with a long term HSHFD,possibly resulting in the improvement on insulin resistance.%目的:观察替米沙坦对长期高糖高脂(HSHF)膳食喂养诱导的胰岛素抵抗 SD大鼠内脏脂肪细胞体积的影响,探讨其对大鼠胰岛素抵抗程度影响的机制。方法将雄性 SD大鼠随机分为3组:正常对照组(对照组,n=10)、高糖高脂组(HSHF组,n=10)、高糖高脂+替米沙坦组(替米沙坦组,n=10)。模型组给予高糖高脂膳食喂养24周后,对照组和 HSHF组继续同前喂养12周,替米沙坦组给予替米沙坦干预12周。试验结束后测定各组大鼠体重、空腹血糖(FPG)、空腹胰岛素(FINS)和三酰甘油(TG),计算胰岛素抵抗指数(HOMA IR);同时留取各组大鼠左侧附睾脂肪组织 HE染色光镜下观察脂肪细胞体积。结果与正常对照组大鼠相比,HSHF组大鼠体重、FPG、FINS、TG、HOMA IR升高,差异有统计学意义(P<0.05);与 HSHF组大鼠相比,替米沙坦组体重、TG、FINS、HOMA IR水平均显著降低,差异有统计学意义(P<0.05)。HE染色镜下观察:正常对照组可见脂肪细胞大小均一,体积小;HSHF组可见脂肪细胞大小不均一,体积较大;相比于 HSHF组,替米沙坦组脂肪细胞体积减小,大小均一性较好。结论替米沙坦能够减轻长期 HSHF膳食喂养大鼠体重,改善其脂代谢,减轻胰岛素抵抗;替米沙坦可减小长期 HSHF膳食喂养大鼠内脏脂肪细胞体积,从而改善胰岛素抵抗。
