慢性乙型肝炎病毒感染者外周血T辅助细胞1、2型因子的表达

摘要

Objective The composition of T helper 0,1,2 cell(Th0/Th1/Th2)in chronic hepatitis B virve(HBV)infected individuals was investigated by determining the expression of IL-4/IFN-γ in a sin-gle CD4+T cell isolate from periphesal blood mononuclear cell(PBMC),and the role of Th1/Th2 cytokines in chronic HBV-infection was analyzed.Methods PBMCs from chronic HBV infected individuals were separated routinely,stimulated by phorboe-12-myristate-13-asetate(PMA)/Ionomycin/Monensin,and the production of interleukin-4(IL-4)interferon-γ(INF-γ)by CD4+T cells in PBMC was determined by fluo-rescence activated call sorting(FAGS)analysis.Results The percentage of IFN-γ-producing T cells and IL-4-producing T cells ranged from 2.3%to 18.6%and from 1.1%to 8.7%respectively in CD4+T cells from non-infected individuals.The majority of CD4+T cells from PBMCs in chronic HBV-infected individ-uals were Th0 cells.The percentage of Th1 cells increased significantly with the increasing of hepatic in-flammation activity.The percentage of Th1 cells in advanced period of chronic hepatitis B was higher than that in stable period of chronic hepatitis B.The percentage of Th2 cells in CD4+T cells from HBV-infected individuals did not differ significantly in different stages,but higher than that of controls.Conclusions The results indicated that Th1 phenotype cytokines were associated with hepatic inflammatory activity of chronic hepatitis B and Th2 cells may be associated with the chronicity of HBV infection.%目的 了解慢性乙型肝炎病毒(HBV)感染者外周血单个核细胞(PBMC)中CD4+T细胞内T辅助细胞1、2型(Thl/Th2)因子、干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)的表达情况,以测定Th0/Thl/Th2细胞的百分数,探明Thl/Th2细胞在慢性HBV感染中的作用.方法 常规分离PBMC,在佛波二酯(PMA)、Ionomycin、Monensin的刺激下,采用流式细胞检测仪(FACS)对慢性HBV感染者外周血单个CD4+T细胞内IL-4和IFN-7表达进行分析.结果 正常对照组,2.3%～18.6%的CD4+T细胞为Th1细胞,0.9%～9.2%为Th2细胞;在慢性HBV感染者外周血单个CD4+T细胞中,以Th0细胞居多;而Th1细胞则随着慢性乙型肝炎肝脏炎症活动的加剧而明显增多,在炎症活动明显的慢性乙型肝炎,其Th1百分数明显高于炎症活动呈静止状态的慢性乙型肝炎;Th2细胞在慢性HBV感染的不同阶段则较为恒定,但明显高于对照组.结论 Th1细胞与炎症活动呈正相关,Th2细胞可能与HBV感染的慢性化相关联.
