Objective To investigate the signaling pathway involved of protective effect during myocardial ischemia-reperfusion (I/R) with resveratrol preconditioning. Methods 48 male SD rats were divided randomly into four groups ( n = 12) , I/R group, resvera-trol pretreatment group( Res + I/R group), resveratrol and sirtl specific inhibitor ex527 pretreatment group ( Res + ex527 + I/R group), ex527 pretreatment group(ex527 + I/R group). The myocardial I/R injury model in rats was established by ligating left anterior descending coronary artery. Spectrophotometry was used to detect the levels of SOD,CK and LDH in the myocardial tissue. TUNEL method was used to detect myocardial cell apoptosis. Bcl-2 and Bax protein were determined by immunohistochemintry, the expressions of sirtl and acetyl-p53 were measured by Western blot Results Resveratrol inhibited myocardial cell apoptosis (P < 0.05) , increased the content of sirtl (P < 0. 05),decreased the acetyl-p53 content(P <0.05) on rat hearts impaired by I/R. Conclusions Preconditioning with resveratrol can inhibit I/R-induced cardiomyocyte apoptosis, the mechanism of which is partly via sirtl -p53 pathway activation.%目的 研究白藜芦醇对大鼠心肌缺血再灌注(I/R)损伤的保护作用及其与sirt1 -p53通路的关系.方法 48只SD大鼠随机分为4组,I/R组、白藜芦醇预处理组(Res+ I/R组),白藜芦醇预处理+sirt1抑制剂组(Res+ ex527+ I/R组),I/R+ sirt1抑制剂组(I/R+ ex527),每组各12只.结扎大鼠冠状动脉左前降支建立大鼠心肌I/R损伤模型;分光光度法测定心肌组织中超氧化物歧化酶(SOD)、肌酸激酶(CK)、乳酸脱氢酶(LDH)含量;TUNEL法检测心肌细胞凋亡;免疫组化测定心肌组织中Bcl-2和Bax蛋白表达;Western印迹检测sirt1、乙酰化p53表达.结果 白藜芦醇能减少大鼠心肌I/R后心肌细胞凋亡(P<0.05),使sirt1表达增高(P<0.05),乙酰化p53表达降低(P<0.05).结论 白藜芦醇对大鼠心肌I/R损伤具有保护作用,其机制可能与sirt1 -p53通路有关.
