Objective To observe the effect of Shenyuheji on kidney cell apoptosis and ERK signal transduction system of diabetic nephropathy (DN) rat. Methods DN SD model rats were made by streptozotocin (STZ) lumbar injection and unilateral nephrectomy and were feed with Shenyuheji gastric perfusion. Cell apoptosis rate and the expressions of caspase-3 and -9 were detected by flow cytometry. The expression of phosphorylation ERK 1/2 protein was measured by Western blotting. Results Shenyuheji could decrease the number of apoptosis cell in DN rat's renal cortex, reduce the expressions of caspase-3 and -9, and activate ERK signal pathway. Conclusions Shenyuheji can efficiently reduce kidney cell apoptosis and activate ERK signal pathway.%目的 观察愈肾合剂对糖尿病(DN)肾病大鼠肾脏细胞凋亡及ERK信号转导系统的影响.方法 SD大鼠单侧肾切除加腹腔注射链脲佐菌素(STZ)复制DN模型,每日愈肾合剂灌胃,8 w后流式细胞术检测肾脏皮质细胞凋亡数目及caspase-9和caspase-3蛋白表达,采用Western 印迹方法检测肾脏皮质细胞内磷酸化ERK1/2蛋白质的表达.结果 愈肾合剂明显减少DN大鼠肾脏皮质细胞凋亡数目,降低caspase-9和caspase-3的蛋白表达,激活ERK信号通路.结论 愈肾合剂可通过抗凋亡途径发挥肾脏保护作用.
