穿心莲内酯对糖尿病大鼠心肌氧化应激的影响

         

摘要

目的 研究穿心莲内酯对实验性糖尿病大鼠心肌氧化应激损伤的影响.方法 SD大鼠高脂高糖饲料喂养4 w后,分两次每次腹腔注射1%的STZ柠檬酸盐缓冲液(STZ 30 mg/kg)制备糖尿病大鼠模型.治疗组分别用2.5%二甲双胍、0.5%和1%的穿心莲内酯10 ml/kg灌胃8w,结束时测定体重指数(LEE指数),心重指数(HW/BW),血糖;心肌组织制备匀浆,比色法测丙二醛(MDA)、超氧化物歧化酶(SOD)、细胞钙含量(CA)、过氧化氢酶(CAT)等指标.结果 穿心莲内酯大剂量组与模型组比较,LEE指数维持正常,心重指数有所下降;心肌组织MDA、CA含量降低,SOD活性升高(P<0.05);穿心莲内酯大剂量组心肌因氧化应激升高的CAT水平降至与正常对照组无异(P>0.05),且与模型组具有显著差异(P<0.05).结论 穿心莲内酯具有减轻糖尿病大鼠心肌氧化应激损伤的作用,并改善大鼠一般情况.%Objective To investigate the effects of andrographolide on oxidative stress of myocardial tissue in rat diabetes model. Methods Following high fat and sugar feed for 4 Weeks, SD rats were injected intraperitoneally with 1% STZ in citrate buffer at 30 mg/kg twice to develop rat diabetes model. The treatment groups were administered intragastriclly with 2. 5% metibrmin, 0. 5% or 1% andrographolide at 10 ml/kg for 8 weeks respectively. At the end of the experiments, body mass index (LEE index) , heart heavy index ( HW/BW) , blood sugar were detected. Malondialdehyde (MDA) , superoxide dismutase (SOD) , cell calcium (CA) , catalase (CAT) in the myocardial tissue homogenate were measured using colorimetric method. Results Compared with model group, LEE index was normal, heart heavy index decreased, MDA, CA content in myocardial tissue were reduced, SOD activity increased in high-dose andrographolide group ■( P < 0. 05). The high level of CAT in high-dose andrographolide group due to oxidative stress fell to the level of normal control group ( P > 0. 05) and there was significant difference {P <0. 05) compared to that of model group. Conclusions Andrographolide could relieve the myocardial oxidative stress damage and improve the general situation in diabetic rats.
