电针通过mTOR/P70 S6 K信号途径增强胰岛素抵抗模型大鼠对胰岛素的敏感性

     

摘要

目的:探讨电针对胰岛素抵抗模型大鼠胰岛素信号转导通路重要影响因子mTOR和 P70S6K mRNA 表达量的影响。方法清洁级SD雄性大鼠70只,按随机数字表分为普食组( n=10)和模型组( n=60),分别饲以普食和高脂高糖饮食。8 w后选取40只造模成功大鼠再一次随机分为高脂高糖饮食(HFHCD)1、HFHCD 2、电针(EA)1、EA 2组,每组10只;HFHCD 1组和EA 1组继续给予高脂高糖饮食,HFHCD 2组和EA 2组给予普通饮食。电针组针刺双侧“足三里”、“三阴交”、“胃脘下俞”,每次20 min,1次/d,针刺14 d。观察大鼠体重、血糖、胰岛素的变化,实时荧光定量PCR检测各组大鼠mTOR和P70S6K mRNA的表达量。结果 HFHCD 1组比CD组mTOR、P70S6KmRNA明显升高,HFHCD 2组比HFHCD 1组明显降低,EA1组较HFHCD1组明显降低,EA2组较HFHCD2组明显降低,EA2组较EA1组明显降低。结论电针刺激胰岛素抵抗大鼠“足三里”、“三阴交”、“胃脘下俞”能降低mTOR和P70S6K mRNA的表达水平,恢复胰岛素信号通路的正常传导,增强胰岛素的敏感性,从而维持血糖的正常水平。%Objective To investigate the effect of electroacupuncture strengthening mechanism of insulin sensitivity on the content of mTORmRNA and P70S6K mRNAin insulin resistance rat.Methods Seventy SD rats were randomly divided into common diet (n=10) and high-fat and high-carbohydrate diet groups(n=60).Forty rats of insulin resistance in high-fat and high-carbohydrate diet group were chosen and divided into HFHCD 1,HFHCD 2,EA 1 and EA 2 groups(n=10).Electroacupuncture was applied to two-side of ST36,SP6 and EX-B3 for 20 min once daily for 14 days.The changes of body weight ,blood glucose and insulin were detected .The expressions of mTOR mRNA and P70S6K mRNA in insulin resis-tance rat skeletal muscle were determined by real time fluorescence quantitative PCR .Results The expres-sions of mTOR and P70S6K mRNA were significantly higher in HFHCD 1 group than those of CD group ,they were significantly lower in HFH-CD 2 group than those of HFHCD 1 group,they were significantly lower in EA1 group than those of HFHCD1 group,they were significantly lower in EA2 group than those of HFHCD2 group,they were significantly lower in EA2 group than those of EA1 group.Conclusions EA on ST36,SP6 and EX-B3 has a beneficial regulatory effect on the expressions of mTOR and P 70S6K mRNA,and restore the normal conduction of insulin singnaling pathway ,and enhance insulin sensitivity ,and maintain the level of blood glucose in normal .
