摘要

Objective To investigate the expressions and clinical significance of miRNAs(miR-150,miR-146a,miR-181a,miR-424,and miR-142-3p) in the peripheral blood mononuclear cells (PBMCs) in patients with type 1 diabetes mellitus (T1DM).Methods 69 patients with T1DM,46 patients with T2DM and 56 healthy individuals were enrolled in this study.The expression levels of miRNAs in the PBMCs were measured using RT-PCR in.ROC curve was used to evaluate the diagnosis value of miRNAs expression in PBMCs for differentiating T1DM from T2DM and healthy controls.The correlation between miRNAs expression and clinical parameters was also analyzed.Results Compared with health control and T2DM group,the expression levels of rniR-150,miR-146a and miR-424 in T1DM patients were decreased[(-1.39±0.49) vs (2.67±0.28) vs (2.95±0.37);(1.08±0.60) vs (4.42±0.87) vs (3.71 ±0.56);(-5.96±0.21) vs (-2.94±0.25) vs (-1.91±0.34);P<0.05].ROC curve analyses revealed that miR-146a,miR-150 and rniR-424 could serve as valuable biomarkers for differentiating T1DM from control group with an AUC (the areas under the ROC curve) of (0.860±0.061),(0.905±0.058)and (0.844±0.066) respectively;rniR-146a,miR-150 and miR-424 could make a distinction between T1DM and T2DM patients with an AUC of (0.868 ± 0.068),(0.921 ± 0.064) and (0.698 ± 0.065),respectively.Compared with GADA autoantibody-negative (Ab-) patients,the expressions of miR-146a,miR-150 and miR-424 were further decreased in PBMCs in GADAb (Ab+) patients[(-2.27± 1.53) vs (-1.11±1.58);(-5.72±1.88) vs (-4.92±1.98);(-0.35±1.61) vs (1.46± 1.99)].Conclusion MiR-150,miR-146a and miR-424 are low expressed in PBMCs in T1DM patients and related with islet autoimmunity which may be helpful for the early detection of T1DM patients.%目的 探讨微小RNA(miR-150、miR-146a、miR-181a、miR-424、miR-142-3p)在T1DM患者外周血单个核细胞(PBMCs)中的表达情况及临床意义.方法 应用实时荧光定量聚合酶反应检测69例T1DM患者、46例T2DM患者和56名健康对照者(NC) PBMCs miRNAs的表达水平.应用受试者工作特征(ROC)曲线评价有差异表达的miRNAs对T1DM的诊断价值,同时将其表达量与临床指标进行相关性分析.结果 与NC组和T2DM组比较,T1DM组PBMCs中miR-146a、miR-424、miR-150相对表达量降低(P<0.05).T1DM组和NC组ROC曲线下面积(AUC)分别为(0.860±0.061)、(0.905±0.058)和(0.844±0.066);T1DM组和T2DM组AUC分别为(0.868±0.068)、(0.921±0.064)和(0.698±0.065);与GADAb抗体阴性者比较,miR-146a、miR-150、miR-424在GADAb抗体阳性的T1DM患者中进一步下降.结论 miR-146a、miR-150、miR-424在T1DM患者PBMCs中低表达,可能与自身免疫状态有关,有助于早期发现T1DM患者.
