辛伐他汀对脂多糖诱导的人脐静脉内皮细胞炎性反应抑制作用

         

摘要

Objective: To observe the effect of simvastatin on the expression of IL-1, IL-6, MCP-1 and uPA/ uPAR mRNA and MCP-1, IL-6 protein in HUVECs induced with LPS. Methods: Cultivated HUVECs were passaged to the third generation. Then they were divided into four groups randomly: the control group, the LPS (100ng/mLgroup), the simvastatin (5 m g/mL) group, the LPS and simvastatin group. The expression of IL-1、IL-6、MCP-1 and uPA/ uPAR mRNA and proteins were measured using real-time quantitative PCR and ELISA. Results: Compared with the control groups.the expression of IL-1、IL-6、MCP-1 and uPA/ uPAR mRNA and proteins in the supernatant were significantly increased in LPS groups(P<0.01). Compared with the LPS groups, the expression of IL-1、L-6、MCP-1 and uPA/ uPAR mRNA and proteins in the supernatant were statistically decreased in LPS and simvastatin groups(P<0.01). Conclusion: Simvastatin can inhibit the synthesis expression of IL-1、IL-6、MCP-1 and uPA/ uPAR in HUVECs,delay the process of atherosclerotic.%目的:运用脂多糖(LPS)诱导体外培养的人脐静脉内皮细胞(HUVECs)的炎性反应,观察辛伐他汀对HUVECs炎性反应的抑制作用.方法:体外培养HUVECs,细胞至第3代,随机分为空白对照组、LPS(100 ng/mL)组、辛伐他汀(5 mg/mL)组、辛伐他汀(5 mg/mL)和LPS (100 ng/mL)共作用组,运用实时荧光定量PCR和ELISA的方法,测定各组细胞单核细胞趋化蛋白-1(MCP-1)、白介素-1(IL-1)、白介素-6(IL-6)和尿激酶型纤溶酶原激活物及其受体(uPA/uPAR)的mRNA和蛋白表达的变化.结果:与对照组相比,LPS组细胞的MCP-1、IL-1、IL-6及uPA/uPAR的mRNA表达和上清液中各因子蛋白的表达均明显增加(P均<0.01);与LPS组相比,辛伐他汀和LPS共作用组的细胞MCP-1、IL-1、IL-6及uPA/uPAR的mRNA表达和上清液中各因子蛋白的表达均明显降低(P均<0.05).结论:辛伐他汀可抑制HUVECs合成及分泌细胞因子MCP-1、IL-1、IL-6及uPA/uPAR,延缓动脉粥样硬化的进程.
