We studied the effects of 15 kinds of hydrophilic cyclodextrins (CyDs) on NO production in several mouse macrophages stimulated with LPS. Among these CyDs, 2,6-di-O-methyl-α-CyD (DM-a-CyD) and 2,6-di-O-methyl-3-O-acetyl-β-cyclodextrin (DMA-β-CyD) significantly inhibited NO production in RAW264.7 cells stimulated with LPS. To gain insight into this inhibition mechanism, the effects of CyDs on the induction of iNOS mRNA and iNOS in RAW264.7 cells as well as on LPS binding to the cell surface and the expression of CD 14 and Toll- like receptor 4 (TLR4)/MD2 complex, sentinel receptors of LPS, on the cell surface were studied.
