Objeetive: To explore the effect of radix isatidis polysaccharide on liver ischemiareperfusion injury (IRI) after liver autotransplantation in rats. Methods: The SD rats were randomly divided into three groups: the autotransplantation group, the radix isatidis polysaccharide + autotransplantation group and the sham operation group, with twenty-four rats in each group. At the 1,12, 24 and 48 hours after operation, the rats were killed and the following tests were conducted: 1)Blood samples were obtained to measure alanine aminotransferase (ALT), aspartate aminotransferase(AST) and tumor necrosis factor α (TNF- α); 2)The morphology of hepatocyte was observed under transmission electron microscopy (TEM). Results The serum levels of ALT, AST and TNF- α in the radix isatidis polysaccharide group were significantly lower than that of the autotransplantation group (P<0.05) after the liver autotransplantation. Under TEM, mitochondria were swollen obviously along with rupture of membrane, some of them even occurred disruption with crista mitochondriales were showed rarefaction or solution in the autotransplantation group. There was no typical change of the structure of mitochondria and the appearance of hepatocytes were normal in the radix isatidis polysaccharide + autotransplantation group. Conclusion: Radix isatidis polysaccharide can protect the hepatocyte after liver transplantation.%目的:探讨板蓝根多糖对自体肝移植大鼠缺血再灌注损伤的作用.方法:将72只SD大鼠随机分为3组:自体肝移植模型组、板蓝根多糖+自体肝移植模型组、假手术组.分别于术后1、12、24、48 h采血检测血清转氨酶及TNF-α的水平,并使用透射电镜观察肝细胞及其线粒体的形态学改变.结果:板蓝根多糖+自体肝移植模型组血清转氨酶及TNF-α水平显著低于自体肝移植模型组,差异有统计学意义(P<0.05).透射电镜示:自体肝移植模型组肝细胞线粒体肿胀明显,膜结构模糊不清、部分破碎,线粒体嵴疏松溶解,有空泡形成;板蓝根多糖+自体肝移植模型组线粒体结构改变不明显,肝细胞结构形态基本正常.结论:板蓝根多糖具有减轻移植肝缺血再灌注损伤的作用.
