黄芩小分子化合物对雌激素缺乏骨质疏松的预防和治疗作用

摘要

AIM:To inviestigate the effects of radices scutellariae extract on the prevention and treatment of oste-oporosis in rats.METHODS:Osteoporosis models of 6 female SD rats were established by ovariectomy .Scutellariae extract was injected intraperitoneally 1 w after ovariectomy in prevention group(n =3),while the extract was given 2 m after ovariectomy in treatment group(n =3).Micro CT scanning of femur was conducted 6 and 12 w after scutellariae extract injection,bone mass and trabecular bone volume were measured.3 rats underwent sham operation were served as the controls.Bone marrow mesenchymal stem cells (BMMSCs)were cultured with or without scutellariae extract , the proliferation and osteogenesis differentiation of BMMSCs were examined.RESULTS:Micro-CT results showed that the bone mass and trabecular bone volume of scutellariae extract group were higher than those of the control group. MTT assay and clone formation assay showed Scutellariae extract promoted BMMSCs proliferation.ALP and Alizarin red staining indicated that scutellariae extract induced osteogenesis differentiation.CONCLUSION:Scutellariae extract have the preventive and treatment effects on osteoporosis,and it can promote the proliferation and osteogenesis differen-tiation of BMMSCs.%目的：探讨黄芩中提取的小分子化合物对大鼠去势后骨质疏松的预防和治疗作用。方法：选取雌性 SD 大鼠建立去势骨质疏松模型，从黄芩中提取小分子化合物，对其腹腔注射。预防组从建模1周开始给药，治疗组从建模2月开始给药，分别在给药后第6、12周未 Micro CT 扫描一侧股骨，比较骨体积和骨小梁数目差异；同时培养各组大鼠骨髓间充质干细胞，检测其对骨髓间充质干细胞增殖和成骨分化的影响。结果：Micro CT 显示，预防组和治疗组动物的骨体积和骨小梁数目相比对照组均有所增加；而 MTT 生长曲线和细胞克隆形成显示药物可以促进培养的骨髓间充质干细胞的增殖；ALP 染色和茜素红染色显示药物对骨髓间充质干细胞的成骨分化也有促进作用。结论：黄芩中小分子化合物可以在一定程度上预防和治疗雌激素缺乏所致的骨质疏松，并促进骨髓间充质干细胞的增殖和成骨分化。
