热休克蛋白27及热休克因子1在牙髓炎中的表达研究

         

摘要

目的:探讨热休克蛋白27(HSP27)和热休克因子1(HSF1)在实验型大鼠牙髓炎模型中的表达.方法:建立实验性大鼠上颌第一、第二磨牙牙髓炎模型并收集临床正常及牙髓炎组织标本,制作连续切片,应用免疫组化方法检测HSP27、HSF1的表达及定位.结果:免疫组化染色发现,大鼠正常牙髓组织中,HSP27阳性表达部位位于成牙本质细胞层;大鼠炎性牙髓组织中HSP27在成牙本质细胞层及炎症牙髓区域均呈阳性表达,且其表达强度随炎症程度逐渐增强;HSF1在大鼠正常及牙髓炎组织中均未表达.人牙髓组织检测结果与动物实验结果相一致.结论:HSP27的表达与牙髓炎的病变程度呈正相关;在牙髓组织损伤修复过程中HSP27可能是介导矿化修复的主要因子之一,而HSF1不参与该修复过程.%AIM:To observe the expression of heat shock protein 27 (HSP27) and heat shock factor 1 (HSF1) in rat dental pulps with experimental pulpitis.METHODS:The pulp chamber of the maxilar 1 st and 2nd molaxs of Wistar rats was opened by a dental bur for the establish ement of experimental pulpitis.The rats were sacrificed on d 1,3,5,7 and 10 after pulp exposure.Maxillary molars with surrounding bone were taken and subjected to immunohistochemical staining for the observation of the expression of HSP27 and HSF1.RESULTS:HSP27 was moderately expressed at the odontoblast layer in normal rat dental tissus.In the middle stage of pulpitis,positive staining of HSP27 was found in odontoblasts,fibroblasts,endothelial cells,smooth muscle cells of the vessel walls and in reparative dentin.The intensity of HSP27 expression increased with the development of pulpitis.However,no HSF1 expression was found in normal and inflammatory pulp tissues.CONCLUSION:HSP27 might be positively related to the development of pulpitis and play a role in the dental pulp repair and in the formation of reparative dentin.HSF1 may have no significance in the progress of pulpitis.
