C6细胞肝与脾原位接种模型的建立及比较

         

摘要

目的 探讨大鼠肝癌细胞系C6肝与脾原位接种成瘤在不同内环境情况下对肿瘤发生与侵袭转移等能力的影响,并筛选最佳C6肝细胞癌肺转移动物模型.方法 C6进行体外培养.将雌性BALB/cA裸鼠随机分为肝与脾原位接种2组.观察裸鼠C6成瘤率、裸鼠存活率、肿瘤病理学特点和远处脏器癌细胞转移等情况.结果 肝原位接种组成瘤率100%,肿瘤呈结节状膨胀性生长,对周围肝组织和腹壁有粘连、侵袭,肺转移率100%.脾原位接种组脾成瘤率100%,肿瘤呈结节状膨胀性生长,对周围组织和腹壁亦有粘连、侵袭,肝、肺转移率分别为100%和71.4%.结论 C6细胞肝和脾原位接种均易成瘤和转移,可作为肝细胞癌(hepatocellular carcinoma,HCC)体内研究和转移模型.脾原位接种模型可能与HCC自发性肿瘤发生和转移的生物学特性更相似,更适合于HCC发生和转移的机制,以及治疗干预等研究.%Purpose To establish and compare the oncogenic and metastatic phenotypes in the mice that their livers or spleens were inoculated with C6 hepatocellular carcinoma cells ( HCC). Methods The HCC C6 cells were cultured in vitro first, and then were injected into liver or spleen in female BALB/cA nude mice. Thereafter, the tumor formation rate, pathologic characteristics, and metastasis were comparatively studied. Results After being injected with C6 cells into their livers, all animals developed a liver tumor. The tumor grew fast and exhibited an expansive growing pattern with surrounding invasion and lung metastasis (100% ). Similarly, after being injected with C6 cells into their spleen, all animals developed splenic tumors, with surrounding invasion, and a high metastatic rate in both liver (100% ) and lung (71.4%). Conclusion Inoculation with C6 cells in the mouse quickly develops hepatocellular cancer in liver and spleen, respectively, with lung metastasis, and this study has thus provided a valuable cancer model in vivo in the mouse. As the direct spleen injection model has more similar carcinogenetic and metastatic features to human HCC, it is probably more useful for the studies of the underlying mechanism and therapeutic regimens for HCC.
