尿毒清颗粒对单侧输尿管梗阻肾间质纤维化大鼠肾脏Toll样受体4表达的影响

摘要

目的 观察尿毒清颗粒对单侧输尿管梗阻模型大鼠肾组织Toll样受体4(TLR4)表达的影响,探讨TLR4在导致肾间质纤维化中的作用.方法 将24只大鼠随机分为正常对照组、假手术组、手术组和尿毒清组,每组6只.14 d后处死大鼠,检测各组大鼠的肾功能及总超氧化物歧化酶(T-SOD)活力、丙二醛(MDA)水平.采用反转录聚合酶链反应(RT-PCR)检测转化生长因子-β1(TGF-β1)mRNA的表达水平.采用免疫组化半定量法分析TLR4表达,并观察肾脏病理变化.结果 4组大鼠尿素氮(BUN)、血肌酐(SCr)、T-SOD、MDA水平比较,差异均有统计学意义(P<0.01=;4组大鼠TGF-β1mRNA和TLR4的表达水平比较,差异均有统计学意义(P<0.01=.其中,假手术组与手术组、手术组与尿毒清组的TGF-β1mRNA和TLR4表达水平比较,差异均有统计学意义(P<0.05).结论 单侧输尿管梗阻可以诱导大鼠肾间质纤维化,氧化应激可能通过TLR4途径参与肾间质纤维化的形成,尿毒清治疗可明显改善大鼠的肾间质纤维化程度.%Objective To explore the changes in toll - like receptor 4 ( TLR4 ) expression in kidneys of unilateral ure-teral obstruction ( UUO ) rats and to investigate the function of toll - like receptor 4 in renal interstitial fibrosis. Methods Twenty - four healthy male SD rats were randomly divided into four groups ( n = 6 ): normal control group, sham group, UUO group, and Niaoduqing treatment group. Rats were sacrificed at day 14 after surgery, with the activity of T - SOD and levels of MDA measured using chemical colorimetry method. Levels of TGF - β1 mRNA were examined by RT - PCR. Pathological changes were examined by HE staining. Toll - like receptor 4 proteins were semi - quantified by immunohistochemistry staining. Results Levels of BUN, SCr, T - SOD, and MDA were significantly different among the four groups ( P <0. 01 ). Levels of TGF -β1 mRNA and TLR4 protein expression was significantly different among the four groups ( P <0. 01 ). Levels of TGF - (3, mRNA and TLR4 protein expression was significantly different between the sham group and the UUO group ( P <0. 05 ). Levels of TGF - p, mRNA and TLR4 protein expression was significantly different between the Niaoduqing group and the UUO group ( P < 0. 05 ). Conclusion Unilateral ureteral obstruction can induce renal interstitial fibrosis in rats. Oxidation stress may contribute to renal interstitial fibrosis via TLR4 signaling. Niaoduqing administration can attenuate renal damage.
