膀胱移行细胞癌中间隙连接蛋白Cx43mRNA、Bcl-2mRNA和Bax mRNA的表达及其相关性研究

摘要

Objective To investigate the expressions of connexin43 , Bcl -2 . and Bax mRNA in bladder transitional cell carcinoma ( BTCC ). Methods The expressions of connexin43 ( Cx43 ), Bcl -2, and Bax mRNA were detected by semi - quantitative RT - PCR in 35 cases of BTCC and 21 cases of para - carcinoma tissue. with 15 cases of normal bladder tissues as controls. The correlation among the expressions of connexin43 . Bcl - 2. and Bax mRNA were explored, and its relationship with the clinical pathology of BTCC were studied. Results Semi - quantitative RT - PCR demonstrated that the expression rate of connexin43 mRNA was ( 0. 34 ±0. 37 ) in BTCC, which was significantly down - regulated compared with that in para - carcinoma tissues ( 0. 79 ±0. 70 ) and normal hladder tissues ( 0. 81 ±0. 47 ) ( P <0. 01 ) . The expression rate of Bcl - 2 mRNA was ( 0. 35±0. 43 ) in BTCC, which was significantly up - regulated when compared with that in para - carcinoma tissues ( 0. 16 ±0. 33 ) and normal bladder tissues ( 0. 15 ±0. 23 ) ( P < 0. 01 ) . No correlation was found between para - carcinoma tissues and normal bladder tissues ( P >0. 05 ) . The expression of Cx43 mRNA was negatively correlated with the grade of the tumor ( P < 0. 05 ),and the expression of Bcl - 2 mRNA showed positive correlation with the grade of the tumor ( P < 0. 05 ) . No correlation was found between the expression of Cx43/ Bcl -2 and the sex, age, clinical stage, and the diameter and the growth pattern of BTCC ( P >0. 05 ); However, the expression of Bcl - 2 mRNA was positively correlated with the recurrence of BTCC. There was no correlation among cancer tissues, para - carcmoma tissues. and normal bladder tissues on the expression of Bax mRNA ( P >0. 05 ) , and no correlation was found hetween the expression of Bax mRNA and the sex, age , grade, clinical stage, and the diameter and the growth pattern of BTCC ( P > 0. 05 ) . Conclusion The down - regulated expression of Cx43 gene is closely associated with the development, invasion. and metastasis of BTCC. Cx43 gene may have antagonistic effects with bcl - 2 gene ( or with bcl - 2/bax ) in the initiation and progression of BTCC. In addition, Cx43 gene may also be involved in the apoptosis of BTCC.%目的 探讨间隙连接蛋白Cx43、Bcl-2及Bax在膀胱移行细胞癌(BTCC)中的表达及其意义.方法 用半定量RT-PCR的方法检测Cx43、Bcl-2及Bax在35例BTCC和21例癌旁组织中的表达情况,并分析其与BTCC临床病理因素的关系.同时取正常膀胱组织15例做对照.结果 BTCC组织中Cx43 mRNA的表达比值[Cx43/人肌动蛋白(β-actin)]为(0.34±0.37),低于BTCC癌旁组织中的(0.79±0.70)和正常膀胱组织的(0.81±0.47),差异有统计学意义(P<0.01);Bcl-2 mRNA表达比值为(0.35±0.43),高于癌旁组织中的(0.16±0.33)和正常膀胱组织的(0.15±0.23),差异有统计学意义(P<0.01).癌旁组织与正常膀胱组织Cx43 mRNA及Bcl-2 mRNA的表达比值比较,差异均无统计学意义(P>0.05).Cx43 mRNA在BTCC病理分级G3的表达为(0.30±0.16),G2的表达为(0.54±0.22),G1的表达为(0.60±0.25),3个级别比较差异有统计学意义(P<0.05).Bcl-2 mRNA在BTCC病理分级G3的表达为(0.47±0.15),G2的表达为(0.35±0.11),G1的表达为(0.24±0.10),3个级别间比较差异有统计学意义(P<0.01).复发BTCC组织中Bcl-2 mRNA的表达水平(0.40±0.15)高于初发癌组织(0.28±0.11),差异有统计学意义(P<0.05).Bax mRNA在BTCC组织、癌旁组织及正常膀胱组织中的表达水平比较,差异无统计学意义(P>0.05).Bax mRNA的表达水平与BTCC患者的肿瘤分期、病理分级以及肿瘤有无复发比较,差异均无统计学意义(P>0.05).结论 Cx43 表达下调与BTCC的发生、进展及其恶性特征有关,Cx43基因可能与凋亡相关基因Bcl-2(或Bcl-2/Bax)在BTCC的发展中起拮抗作用,与细胞的凋亡过程有关.
