加味桂枝茯苓颗粒对大鼠前列腺增生组织血管内皮生长因子和碱性成纤维细胞生长因子表达水平的影响

摘要

Objective To explore the mechanism of flavored cassia twig tuckahoe particles in the treatment of benign prostatic hyperplasia(BPH)by researching the influence of flavored cassia twig tuckahoe particles on the expression of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF). Methods From January 2013 to June 2015,we selected 90 male SD rats. Using random number table method,we divided the rats into normal group(group A,n = 25)and model group(n = 65). After model building,we randomly selected 20 rats from group A and 60 rats from model group. The 60 rats selected from model group were further divided into three groups:control group( group B,n = 20),flavored cassia twig tuckahoe particles group(group C,n = 20)and finasteride group(group D,n = 20)by randorn number table method. The rats of group A were normally fed;group B was given gavage with sodium chloride solution;group C was given gavage with flavored cassia twig tuckahoe particle solution;group D was given gavage with finasteride solution. Twenty - four hours after last gavage, prostate tissues were sampled,and we recorded the prostate volume and prostate hyperplasia index of each group;the expression levels of VEGF and bFGF were detected by immunohistochemical SP method. Results Group B was higher than group A in prostate volume and prostate hyperplasia index(P < 0. 05);group C and group D were lower than group B in prostate volume and prostate hyperplasia index(P < 0. 05). Group B,group C and group D were higher than group A in the levels of VEGF and bFGF(P < 0. 05);group C and group D were lower than group B in the levels of VEGF and bFGF(P < 0. 05). Conclusion Flavored cassia twig tuckahoe particles can lessen prostate volume and prostate hyperplasia index by reducing the levels of VEGF and bFGF in the hyperplasia of prostate gland tissue of rats,thus achieving efficacy in BPH treatment.%目的:通过加味桂枝茯苓颗粒对大鼠前列腺增生组织中血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)表达水平影响的研究,探讨加味桂枝茯苓颗粒对良性前列腺增生症( BPH)的作用机制。方法2013年1月—2015年6月,选取90只雄性 SD 大鼠,采用随机数字表法将其分成正常组( A 组,25只)和模型组(65只),A 组随机取20只,模型组在验证造模成功后随机取60只,模型组采用随机数字表进一步分为对照组( B组,20只)、加味桂枝茯苓颗粒组(C 组,20只)、非那雄胺片组(D 组,20只)。A 组大鼠正常饲养,模型组造模,之后 B 组大鼠0.9%氯化钠溶液灌胃,C 组大鼠加味桂枝茯苓颗粒溶液灌胃,D 组大鼠非那雄胺片溶液灌胃。于末次灌胃治疗结束24 h 后进行前列腺组织标本采集。记录各组大鼠前列腺体积、前列腺增生指数,采用免疫组化 SP 法检测VEGF、bFGF 表达水平。结果 B 组前列腺体积、前列腺增生指数大于 A 组(P <0.05);C 组、D 组前列腺体积、前列腺增生指数小于 B 组( P <0.05)。B 组、C 组、D 组 VEGF、bFGF 表达水平高于 A 组( P <0.05);C 组、D 组VEGF、bFGF 表达水平低于 B 组(P <0.05)。结论加味桂枝茯苓颗粒通过降低大鼠前列腺增生腺体组织中 VEGF、bFGF 表达水平,进而缩小前列腺体积,降低前列腺增生指数,从而达到治疗 BPH 的效果。
