膀胱尿路上皮癌标本中人微小核糖核酸3658与人源性长寿保障基因Ⅱ型基因表达的相关性及其临床意义

摘要

目的:探讨人微小核糖核酸3658(miR-3658)和人源性长寿保障基因Ⅱ型(LASS2)在临床膀胱尿路上皮癌标本中的表达差异及相关性。方法收集昆明医科大学第二附属医院和云南省肿瘤医院泌尿外科2014年1月—2015年1月收治的膀胱癌患者96例,在膀胱癌根治术中获取膀胱癌组织和癌旁组织。采用实时荧光定量 PCR 测定膀胱癌组织和癌旁组织miR-3658及 LASS2 mRNA 表达水平;采用免疫组化法检测膀胱癌组织和癌旁组织 LASS2蛋白表达情况。结果膀胱癌组织miR-3658 mRNA 表达水平高于癌旁组织,LASS2 mRNA 表达水平低于癌旁组织,差异均有统计学意义( t =6.46、38.12,P ﹤0.01)。不同复发性、病理分级、浸润深度、淋巴转移、远处转移、TNM 分期者miR-3658、LASS2 mRNA 表达水平比较,差异均有统计学意义(P ﹤0.05)。LASS2蛋白表达定位于胞质中,呈棕色或黄褐色颗粒状。膀胱癌组织 LASS2蛋白阳性表达率低于癌旁组织,差异有统计学意义(χ2=15.32,P ﹤0.01)。不同复发性、病理分级、浸润深度、淋巴转移、TNM 分期者 LASS2蛋白阳性表达率比较,差异均有统计学意义( P ﹤0.05)。Pearson 相关分析结果显示,膀胱癌组织 LASS2 mRNA 表达水平与miR-3658 mRNA 表达水平呈负相关( r =-0.676,P ﹤0.05)。结论 miR-3658与 LASS2在膀胱癌组织中的表达呈负相关,miR-3658可能通过调节 LASS2表达而参与膀胱癌发生发展的过程。%Objective To investigate the expression differences and correlation of microRNA - 3658( miR-3658) and homo sapiens longevity assurance homologue 2(LASS2) in clinical bladder urothelial carcinoma samples. Methods We collected 96 patients with bladder urothelial carcinoma from the Departments of Urology in the Second Affiliated Hospital of Kunming Medical University and Yunnan Cancer Hospital from January 2014 to January 2015;bladder cancer tissues and paracarcinoma tissues were obtained from radical resection of bladder urothelial carcinoma. Hsa-miR-3658 and LASS2 mRNA expression in bladder cancer tissues and paracarcinoma tissues were detected by real - time fluorescence quantification PCR;LASS2 protein expression in bladder cancer tissues and paracarcinoma tissues was detected by immunohistochemistry( IHC). Results miR-3658 mRNA expression was higher in bladder cancer tissues than paracarcinoma tissues;and LASS2 mRNA expression was lower in bladder cancer tissues than paracarcinoma tissues;the two differences were both significant(t = 6. 46, 38. 12;P ﹤ 0. 01). miR-3658 mRNA and LASS2 mRNA expression were significantly different among patients with different recurrence status,pathological grade,infiltration depth,lymphatic metastasis,distal metastasis and TNM stages(P ﹤ 0. 05). LASS2 protein located in cytoplasm was brown or yellow - brown granules. The positive expression rate of LASS2 protein was lower in bladder cancer tissues than paracarcinoma tissues( χ2 = 15. 32,P ﹤ 0. 01). The positive expression rate of LASS2 protein was significantly different among patients with different recurrence status,pathological grades, infiltration depth, lymphatic metastasis and TNM stages( P ﹤ 0. 05). Pearson correlation analysis results showed LASS2 mRNA and miR-3658 mRNA expression were negatively correlated in bladder cancer tissues( r = - 0. 676,P ﹤ 0. 05). Conclusion miR-3658 expression is negatively correlated with LASS2 expression in bladder cancer tissues,and miR-3658 may participate in the occurrence and development of bladder cancer by regulating the LASS2 expression.
