研究发现脂肪酸合成酶(fatty acid synthase,FAS)是一种可能的癌基因,在一般正常组织中表达较低,但在多种恶性肿瘤中表达较高,且FAS的高表达与肿瘤的发生、发展以及预后密切相关。近年来大量研究表明,FAS抑制剂具有明显的抗肿瘤作用,其机制与诱导肿瘤细胞凋亡、抑制肿瘤细胞增殖、抑制肿瘤侵袭及转移等有关,同时FAS抑制剂也具有增敏化疗药物的作用。本文对近年来FAS抑制剂抗肿瘤作用机制进行综述,对其抗肿瘤作用的研究方向进行展望。%It has been proved that FAS is a potent oncogene. Its expression and activity is normal in most of the non-neoplastic cells, but it is over-expressed and associated with poor prognosis in several human malignancies. Recent studies show that FAS inhibitor can induce significant anti-tumor effect. It reduces cell proliferation,enhances apopto-sis,inhibits tumor invasion and metastasis,while it is also an chemosensitization for other chemotherapy drugs. In this paper, the anti-tumor mechanism of FAS inhibitors is reviewed to prospect the research direction of its anti-tumor ef-fect.
