苦参碱改构体X抑制鼻咽癌细胞CNE2增殖作用的实验研究

     

摘要

Objective: To study effects and mechanism of modification X of the Matrine on inhibiting the growth of human nasopharyngeal carcinoma CNE2 cells. Methods: In this study, the author divided it into five groups that the experiment group, negative control group, normal cell control group, solvent group and blank control group (zero hole); cell morphology were observed by inverted optical microscope, cell suppression rates were measured by the MTT assay, cell cycle and apoptosis were measured hy flow cytometry. Results: Dosing cells were round and smaller, and there were some vacuoles in the dosing cells; inhibition of the body were clear and demonstrated concentration-dependent (P<0.05), and it's inhihition rate went up to 97.9%, but no clear inhibition on the human umbilical vein endothelial cells and it's inhibition rate were only up to 7.7%; cell cycle of CNE2 arrested at the S cycle and the apoptosis were clear and concentration-dependent, the apoptosis rate were up to 89.71%. Conclusion: Modification X of the Matrine would arrest the S cycle and induce apoptosis to inhibit the growth of CNE2 Cells.%目的:探讨苦参碱改构体X对人鼻咽癌细胞株CNE2的增殖抑制作用及其可能的机制.方法:本实验分5组,即实验组、阴性对照组、正常细胞对照组、溶剂组、空白对照组(调零孔);倒置光学显微镜下观察细胞形态:采用MTT实验检测药物抑制率;流式细胞仪检测细胞周期的分布和凋亡情况.结果:加药细胞组形态变圆、变小、胞内可见空泡;MTT检测可见药物对人鼻咽癌细胞株CNE2有显著的抑制作用(P<0.05),抑制率最大可达到97.9%,但是对人脐静脉内皮细胞没有表现出相应的抑制作用,抑制率最大仅达到7.7%;流式细胞仪检测结果可见CNE2细胞的周期阻滞在S期和有浓度依赖性的明显凋亡,凋亡率最大可达89.71%.结论:苦参碱改构体X可通过细胞周期S期阻滞和诱导细胞凋亡来抑制鼻咽癌细胞的增殖分裂.
