基质金属蛋白酶-7、上皮性钙黏附蛋白和血管内皮生长因子在乳腺癌中表达及意义的研究

         

摘要

目的 探讨基质金属蛋白酶-7(MMP-7)、上皮性钙黏附蛋白(E-cad)和血管内皮生长因子(VEGF)在乳腺癌中的表达及意义,以期进一步阐明乳腺癌浸润、转移的机制.方法 乳腺原发性浸润性导管癌术后存档蜡块标本68例,导管原位癌25例,并随机选取正常乳腺组织17例作为对照.将所有石蜡包埋标本制成组织芯片后应用免疫组织化学方法检测三种蛋白的表达情况.结果 乳腺导管原位癌和浸润性导管癌MMP-7、VEGF表达均明显高于正常乳腺组织,而E-cad表达明显低于正常乳腺组织,乳腺导管原位癌和浸润性导管癌MMP-7、E-cad和VEGF的表达均没有差异.乳腺导管癌中MMP-7 、E-cad和VEGF的表达与肿瘤分化程度、临床分期及淋巴结转移有关.结论 MMP-7、E-cad和VEGF三者可能均参与了乳腺癌发生发展过程.%Objective To investigate the expression and significance of MMP-7, E-cad and VEGF, and further explore the possible mechanism of carcinogenesis and progression of human ductal breast carcinoma. Methods Immunohistochemistry was used to detect the expression of MMP-7, E-cad and VEGF protein in 68 cases of infiltrating ductal carcinoma, 25 cases of ductal carcinoma in situ and 17 cases of normal breast tissue. Results The expressions of MMP-7 and VEGF in infiltrating ductal carcinoma and ductal carcinoma in situ were all significantly higher than those in normal breast tissue, while the expression of E-cad was significantly lower. No significant difference was detected in the expression of MMP-7, VEGF and E-cad between infiltrating ductal carcinoma and ductal carcinoma in situ. The expressions of MMP-7, E-cad and VEGF in breast tissue of ductal carcinoma were associated with differentiation, TNM stage and lymphatic metastasis. Conclusion MMP-7, E-cad and VEGF may all be involved in the carcinogenesis and progression of breast ductal carcinoma.
