同源异形盒A10及上皮性钙黏蛋白在人子宫内膜癌组织中的表达

         

摘要

Objective To explore the expression and correlation of homeobox gene A10 (HOXA10) and E-cadherin (E-cad) in endometrial carcinoma, and to investigate the effect of HOXA10 and E-cad during the development and metas-tasis of endometrial carcinoma. Methods Paraffin blocks on file removed by surgery from 40 women with endometrial carcinoma in Jinzhou Central Hospital from January 2012 to January 2013 were selected as experimental subject, and normal endometrium specimens from 30 women taking hysterectomy because of benign lesion were selected as controls. The expression of HOXA10 and E-cad proteins in endometrial carcinoma and normal endometrium was detected by im-munohistochemical technique. Results The positive expression rate of HOXA10 protein in endometrial carcinoma was obviously lower, the normal expression rate of E-cad protein in endometrial carcinoma was obviously lower, which had significant differences compared with normal endometrium (P< 0.05); the expression of HOXA10 and E-cad proteins in endometrial carcinoma was correlated with histological grade, clinical stages. depth of myometrial invasion, whether there was lymphatic metastasis (P< 0.05); both showed moderate positive correlation in the expression of endometrial carcinoma (r=0.448, P<0.05). Conclusion The declined or missing expression of HOXA10 in endometrial carcinoma may lower the expression of E-cad through certain mechanisms, and promote the metastasis and invasion of tumor through the transformed way of epithelial and stromal. The combined detection of the expression of HOXA10 and E-cad proteins in endometrial carcinoma may help to understand the grade malignancy of endometrial carcinoma.%目的:探讨同源异型盒基因A10(HOXA10)及上皮性钙黏蛋白(E-cad)在人子宫内膜癌组织中的表达及其相关性,探讨二者在子宫内膜癌发生发展及浸润转移中的作用。方法选取锦州市中心医院2012年1月~2013年1月手术切除的40例子宫内膜癌存档蜡块作为实验对象,选择同期因良性病变切除子宫的正常子宫内膜标本30例作为对照。免疫组化实验检测子宫内膜癌和正常子宫内膜标本中HOXA10和E-cad蛋白的表达。结果子宫内膜癌中HOXA10蛋白的阳性表达率低,E-cad蛋白的正常表达率低,与正常子宫内膜比较,差异有统计学意义(P<0.05);并与子宫内膜癌的组织学分级、临床分期、肌层浸润深度、有无淋巴结转移有关(P<0.05);二者在子宫内膜癌中的表达呈中度正相关(r=0.448,P<0.05)。结论子宫内膜癌组织HOXA10表达下降或缺失可能通过某种机制下调了E-cad的表达,通过上皮间质转化途径促进了肿瘤的转移和侵袭。联合检测HOXA10和E-cad在子宫内膜癌的表达,可能有助于了解子宫内膜癌的恶性程度。
