内皮抑制素对皮肤基底细胞癌 A431细胞株耐药基因的影响

     

摘要

Objective:To assess the effect of endostatin on drug resistance genes MDR1 and GST-πof bas ̄al cell carcinoma A431 cells. Methods: The proliferation of A431 cells was measured with MTT assay. The expression of MDR1 and GST-π were determined by reverse transcription-polymerase chain reaction (RT-PCR) and the expression of MDR1 and GST-πprotein were determined by Western blot in basal cell carcino ̄ma A431 cells. Results:The proliferation of A431 cells was inhibited by endostatin in a time-dose dependent manner. The expression of mRNA and protein of MDR1、GST-π was significantly decreased in the endostatin group than in control group (P<0.01). Conclusion: Endostatin can restore the sensitivity of A431 cells to chemotherapeutic drugs by regulating the expression of MDR1 and GST-π.%  目的:评价内皮抑制素对人皮肤基底细胞癌A431细胞耐药基因MDR1、GST-π的影响。方法:四甲基偶氮唑蓝(MTT)法测定内皮抑制素对A431细胞的增殖抑制率;反转录-聚合酶链反应法(RT-PCR)检测细胞多药耐药基因1(MDR1)、GST-πmRNA的表达水平;蛋白印迹(Western blot)法检测细胞MDR1和GST-π蛋白表达情况。结果:内皮抑制素对A431细胞的增殖抑制率呈时间-剂量依赖性,而内皮抑制素组MDR1、GST-π mRNA及蛋白表达水平均明显低于对照组(P<0.01)。结论:内皮抑制素通过调节耐药基因MDR1、GST-π的表达可能有助于A431细胞恢复对化疗药物的敏感性。
