TGF-β1调节瘢痕疙瘩成纤维细胞分泌MMP-1及TIMP-1的研究

         

摘要

目的:检测转化生长因子-β1(TGF-β1)对瘢痕疙瘩成纤维细胞分泌基质金属蛋白酶-1(MMP-1)和金属蛋白酶组织抑制剂-1(TIMP-1)表达的调节作用,探讨TGF-β1与瘢痕的关系。方法:培养的瘢痕疙瘩成纤维细胞经不同浓度的TGF-β1处理,采用酶联免疫法(ELISA)测定瘢痕疙瘩成纤维细胞上清液中MMP-1和TIMP-1的水平。结果:瘢痕疙瘩中成纤维细胞MMP-1和TIMP-1的生成量显著高于正常皮肤( P<0.01);TGF-β1显著降低瘢痕成纤维细胞MMP-1的生成量( P<0.01),其作用在10~50 ng?mL呈剂量效应关系;不同浓度的TGF-β1对TIMP-1的生成量均无显著改变(P>0.05)。结论: TGF-β1在瘢痕生成过程中起重要作用。%  Objective: To investigate the regulatory effects of transforming growth factor-β1 (TGF-β1) on the secretion of matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in hu-man keloid- derived fibroblasts, and to understand the relationship between TGF-β1 and the scar formation. Method: The cultured keloid fibroblasts were treated with TGF-β1 at different concentrations. Enzyme linked im-munosorbent assay (ELISA) method were performed to measure the expression of MMP-1 and TIMP-1 in human keloid fibroblasts supernatant. Results:The expression of MMP-1 and TIMP-1 was significantly higher in human keloid fibroblasts than in normal dermal fibroblasts (P<0.01). The expression of MMP-1 was decreased signifi-cantly after TGF-β1 was added into the cultured keloid fibroblasts (P<0.01), with a dose-dependent as the in-creasing at concentrations of 10~50 ng?mL. However, the adding of TGF-β1 did not have any influence on the ex-pression of TIMP-1 (P>0.05). Conclusion: TGF-β1 may play a role in the regulation of keloid formation.
