目的:探究不同分子生物学方法检测手足口病标本的应用。方法选取该院2014年2月-2015年9月采集的209例手足口病标本作为对象进行研究,分别予以巢式PCR、RT-PCR检测,比较两种检测方法检测结果。结果巢式PCR检测CA16阳性率、EV71阳性率及CA16+EV71双阳性率均显著高于RT-PCR检测,差异有统计学意义(P﹤0.05);对比Agilent2100与普通凝胶电泳结果,差异无统计学意义(P>0.05);巢式PCR检测对粪便标本、咽拭子标本的检测灵敏度均显著高于RT-PCR检测,差异有统计学意义(P﹤0.05)。结论应用巢式PCR检测手足口病标本,其检测灵敏度较高。%Objective To study the application of different molecular biology methods in detecting hand-foot-mouth dis-ease. Methods 209 cases of hand-foot-mouth specimens collected in our hospital from February 2014 to September 2015 were selected as the research objects, and respectively detected by nested PCR and RT-PCR detection, and the detection results of the two methods were compared. Results The positive rates of nested PCR in diagnosis of CA16, EV71 and CA16+EV71 were obviously higher than those of RT-PCR detection, and the differences had statistical significance, P﹤0.05, and the difference between the Agilent 2100 result and common gel electrophoresis result had no statistical signifi-cance, P>0.05, and the detection sensitivity degree of nested PCR for stool specimens and pharynx specimens was obviously higher than that of RT-PCR detection and the difference had statistical significance, P﹤0.05. Conclusion The application of nested PCR in detecting hand-foot-mouth disease specimens has a higher detection sensitivity degree.
