狼疮肾炎患者循环miRNA表达谱研究及循环miR-130b-3p的临床意义

pdf Objective To study the clinical expression and significance of circulating miRNA and provide more objective basis in the treatment of LN (Lupus Nephritis). Methods Convenient selection 50 LN patients treated from September 2013 to September 2015 in our hospital were selected. 50 healthy check-ups were selected as control group. 15 subjects were randomly selected to receive the detection of circulating miRNA expression profiling (detection group); 85 subjects received the verification of circulating miRNA expression profiling (verification group); at the same time, the circulating miR-130b-3p was the verification object; Observation and comparison of lupus nephritis patients and the control group of patients and the serum levels of total RNA and detecting group in two groups of patients with serum miRNA levels, the validation group miR-130b-3p level, and data statistical analysis and correlation analysis Results In the validation group, and control group comparison (3.7± 1.6)ng/μL, significantly increased the expression levels of miR-130b-3p of circulating serum in patients with lupus nephritis (9.8±1.4)ng/μL, the difference has statistical significance(P<0.05). In the test group, compared with the control group (28.6±2.4)ng/μL, miRNA levels (46.8±5.6)μg/uL were significantly increased in patients with lupus nephritis, and the difference was statistically significant (P< 0.05). Conclusion For LN patients, the expression of circulating miRNA may be degraded with the lower renal function. The expression of circulating miR-130b-3p has an obvious correlation with renal function and blood lipid level, which it plays an important role in the genesis of LN.%目的：研究探讨狼疮肾炎患者外周循环miRNA表达变化及其可能存在的临床意义,为狼疮肾炎的临床治疗提供更多的客观依据。方法方便选取2013年9月-2015年9月于该院诊治的50例狼疮肾炎患者,同时另选同期于该院体检的50名健康者作为对照组。在100例研究对象中随机选取15例患者做循环miRNA的表达谱检测筛选(检测组),85例患者做循环miRNA的表达谱验证筛选(验证组),同时应用循环miR-130b-3p为验证对象,观察与比较狼疮肾炎患者与对照组患者的血清总RNA水平及检测组中两组患者的血清miRNA水平、验证组中miR-130b-3p的水平,并进行数据统计学分析及相关性分析。结果在验证组中,与对照组相比较(3.7±1.6)ng/μL,狼疮肾炎组患者的循环血清中miR-130b-3p的表达水平(9.8±1.4)ng/μL显著上升,其差异有统计学意义(P<0.05)；在检测组中,与对照组相比较(28.6±2.4)ng/μL,狼疮肾炎组患者循环表达的miRNA水平(46.8±5.6)ng/μL明显上升,其差异具有统计学意义(P<0.05)。结论狼疮肾炎患者循环miRNA表达水平可能随着肾功能的进一步下降而出现表达下调,其循环miR-130b-3p的表达与肾脏功能、血脂水平呈现明显的相关性,这在狼疮肾炎的疾病发生发展中可能起着重要的作用。
