三倍体无籽西瓜子叶节再生体系建立与嫁接育苗技术探究

         

摘要

In this paper,we used seedless watermelon ‘ Chaoting No.3’ cotyledonary node as explants for the establishment of seedless watermelon cotyledon node regeneration and grafting breeding technology system to investigate the key factors affecting cotyledon node regeneration in vitro and the grafting seedling techniques.The result showed that 3% of sodium hypochlorite solution of 18 min was the best disinfection technique to seedless embryo.2.5 mg· L-1 6-BA+0.1 mg·L-1 NAA had the best adventitious bud induction in seedless watermelon,the differentiation coefficient of adventitious bud was as high as 6.11.0.2 mg· L-1 KT could effectively induce adventitious bud to elongate.Scions with 2.5 cm in length which were grated to pumpkin root stocks with flatted cotyledon had the the highest survival rate (84.4%).In the experiment,we successfully established the system of cotyledon node regeneration and vitro-grafting technology in triploid seedless watermelon.%以三倍体无籽西瓜品种‘超庭三号’子叶节为外植体,对影响子叶节离体再生及嫁接育苗技术的关键因子进行了研究.结果表明,3%的次氯酸钠溶液消毒18 min是三倍体无籽西瓜种胚消毒的最佳方式;2.5 mg·L-16-BA+0.1 mg·L-1 NAA是诱导三倍体无籽西瓜子叶节产生不定芽的最佳激素组合,不定芽分化系数最高达6.11;0.2 mg·L-1KT能有效诱导不定芽伸长;将长度为2.5 cm的接穗嫁接到子叶刚展平的南瓜砧木上,成活率高达84.40%.试验成功建立了三倍体无籽西瓜子叶节再生及嫁接育苗技术体系.
