To establish a method for detecting aflatoxins content in soy sauce mash by high performance liquid chromatography equipped with fluorescence detection (HPLC-FLD).Method:The HPLC-FLD method used for detecting AFT in soy sauce mash is established based on the volumetric optimization of derivatizing agent (trifluoroacetic acid)and cleaning up with immunoaffinity columns, by which,AFT in ten kinds of soy sauce mash samples is detected.Result:The limitation of quantity of aflatoxins B1 ,B2 ,G1 ,G2 is 0.32,0.35,1.10,0.35 ng/mL respectively,and four kinds of aflatoxins in 0.5~50 ng/mL have an excellent linear relationship.The detection results of aflatoxins in different types of samples show that AFB1 ,AFB2 ,AFG1 ,AFG2 could be detected by this method, which is favorable to eliminate the interference of contaminants such as fats and proteins.Conclusion:The method established has high sensitivity and is simple and fast,which is suitable for determining AFT in soy sauce mash.%目的:建立了一种基于柱前衍生的高效液相色谱-荧光检测法(HPLC-FLD)检测酱醪中黄曲霉毒素(AFT)的方法。方法:通过利用免疫亲和柱净化后,经三氟乙酸(TFA)柱前衍生,建立了HPLC-FLD检测酱醪中AFT 的方法,并对10种酱醪样品中 AFT 的含量进行了检测。结果:该方法中 AFB1, AFB2,AFG1,AFG2的定量检测限(LOQ)分别为0.32,0.35,1.10,0.35 ng/mL,且这4种 AFT 在0.5~50 ng/mL之间线性关系良好,方法稳定。不同类型样品的检测结果表明:该方法可有效克服样品中脂肪、蛋白质等杂质的干扰,同时可用于检测AFB1,AFB2,AFG1,AFG24种黄曲霉毒素。结论:该方法灵敏度高,简单快捷,适用于酱醪中AFT的检测。
