在摇瓶发酵条件下,采用单因素实验和响应面分析法对枯草芽孢杆菌ZC1的发酵培养基和条件进行优化,以提高其活菌数。通过单因素实验和响应面分析,确定了枯草芽孢杆菌ZC1的最佳发酵培养基:豆粕粉24 g/L、葡萄糖6 g/L、氯化钠8 g/L;发酵条件:发酵温度37℃、初始pH 7.5、接种量4%、摇床转速200 r/min。在此条件下,发酵24 h,枯草芽孢杆菌 ZC1的活菌数由原来的1.13×1010 cfu/mL提高到3.69×1010 cfu/mL。%By shake flask fermentation,in order to increase the viable count of Bacillus subtilis ZC1 , the fermentation medium and conditions of Bacillus subtilis ZC1 are optimized by single factor experiment and response surface methodology.According to the single factor experiment and response surface methodology,the optimal fermentation medium and conditions are as follows:soybean meal is 24 g/L,glucose is 6 g/L,sodium chloride is 8 g/L,fermentation temperature is 37 ℃,initial pH is 7 .5 ,inoculation amount is 4%,shaking speed is 200 r/min.Being fermented for 24 h,the viable count of Bacillussubtilis ZC1 is increased from 1.13×1010 cfu/mL to 3.69×1010 cfu/mL under such conditions .
