表达猪流行性腹泻病毒保护性抗原重组乳酸杆菌的构建及鉴定

             

摘要

In order to develop a safe and effective oral vaccine for Porcine epidemic diarrhea(PED),the core neutralizing epitope(COE)gene of PEDV was cloned and inserted into the shuttle expression vector. Then the vector was electric-transformed into the genetically engineered strain ofLactobacilluscasei. After bacteria culture and protein induction,a stripe of about 27 KD was detected by western blot. As a result,the develop of recombinantLactobacillus laid foundation for further study of animal immunological experiments in vivo and novel PED mucosal vaccine.%为开发一种安全有效的新型猪流行性腹泻口服疫苗,将猪流行性腹泻病毒核心保护性抗原基因(COE)插入到穿梭表达载体,并电转化至乳酸杆菌基因工程菌株内,进行诱导培养,通过Western blot能够检测到大小约27 KD的蛋白表达.该重组乳酸杆菌的构建为动物体内免疫试验的开展和新型黏膜免疫疫苗的研发奠定了基础.
