奥曲肽对实验性肝纤维化大鼠瘦素及功能性瘦素受体表达的影响

         

摘要

目的 探讨奥曲肽(OCT)对实验性肝纤维化大鼠瘦素及功能性瘦素受体(OB-Rb)表达的影响.方法 雄性SD大鼠54只,随机分为3组:正常组、模型组、OCT组.采用四氯化碳(CCl4)复合因素建立肝纤维化模型,实验开始后分别于第2、4、6周随机采集标本,测定血清肝功能、瘦素及转化生长因子β1(TGF-β1)水平,观察肝脏病理形态学改变,检测肝纤维化大鼠肝组织瘦素及OB-Rb的表达情况.结果 与正常组比较,模型组和OCT组大鼠血清肝功能下降,瘦素及TGF-β1水平增高,肝组织瘦素及OB-Rb表达增强,且随着时间的增加表现明显;OCT组与模型组同时相比较,OCT组肝功能下降程度、瘦素及TGF-β1水平增高程度较小,病理改变较轻,肝组织瘦素及OB-Rb的表达水平较低.结论 OCT可在一定程度上改善肝功能,延缓肝纤维化的进展,其机制可能是OCT通过抑制瘦素及OB-Rb的表达而影响TGF-β1水平,进而延缓肝纤维化的进展.%Objective To explore the effects of octreotide ( OCT ) on expressions of leptin and obese receptor ( OB-Rb ) of experimental liver fibrosis in rats. Methods 54 adult male SD rats were randomly divided into three groups: normal group, model group and OCT treatment group. The liver fibrosis models in rats were established by carbon tetrachloride ( CC14 ) with compound factors. OCT with abdominal subcutaneous injection was administered twice a day in OCT group. Three groups of rats were sacrificed randomly on the 2,4,6 weeks respectively. The levels of serum total bilirubin ( TBIL ), alanine aminotransferase ( ALT ), aspartate aminotransferase ( AST ), albumin ( ALB ), leptin and transforming growth factor betal( TGF-β1 ) were detected and the histopathology changes were observed; The immunohistochemical technique was used for detecting the leptin and OB-Rb protein expression. Results With the experimental time extending, the levels of serum TBIL, ALT, AST, leptin and TGF-β1 were increased, model group and OCT group were higher than those of normal group; The leptin and OB-Rb protein expressions increased. Compared with model group, the injury degree of hepatic function and histopathology of changes in OCT group decreased, the leptin and OB-Rb protein expression decreased. Conclusion OCT can improve the degree of hepatic function injury, delay the progress of liver fibrosis, which may be related with reducing the levels of TGF-β1 by inhibiting the expression of leptin and OB-Rb.
