取90 g SPF级SD大鼠的双后肢骨髓,分别使用直接贴壁法和密度梯度离心法获得大鼠骨髓单个核细胞,用内皮培养体系EGM-2培养基进行定向诱导分化7~14 d获得大鼠内皮祖细胞。免疫荧光染色和细胞功能学鉴定内皮祖细胞,观察内皮祖细胞出现的时间和数量, CCK-8法鉴定细胞的增殖能力。与密度梯度离心法相比,直接贴壁法在内皮祖细胞出现的时间、数量和增殖能力差异有统计学意义(P<0.01)。因此,相对于密度梯度离心法,直接贴壁法培养出的内皮祖细胞分化时间更短、数量更多且增殖能力更强。%The bone marrow taken from both hindlimbs of SD rats was divided into two parts, mononuclear cell was obtained by direct adherent method and density gradient centrifugation, respectively. Endothelial progenitor cell (EPC) was induced to differentiate from mononuclear cell when cultured 7 to 14 days in EGM-2. EPC was identi-fied by immunofluorescence staining and experimental study of cell function; number of EPC and the time of EPC appeared were recorded;proliferation ability was examined by CCK-8 method. The time when EPC first emerged, number of EPC, and proliferation ability in direct adherent group were significantly than those in density gradient centrifugation group. Therefore, compared with density gradient centrifugation, the cell isolated by direct adherent method could get more cells, need less time to differentiate into EPC and have bigger proliferation potential.
