丹参多酚酸盐抑制培养大鼠系膜细胞增殖及内皮素释放

         

摘要

AIM: To study the effects of salvianolate, an aqueous rnextract of Radix Salviae Miltiorrhizae, on the proliferation rnand endothelin release of cultured rat mesangial cells.rnMETHODS: The proliferation of mesangial cells was rndetermined in terms of [3H]thymidine uptake. The conrncentration of endothelin was measured by radioimmunoasrnsay. The cytotoxicity of salvianolate was tested by tetrarnzolium (MTT) and lactic dehydrogenase (LDH) assay.rnRESULTS: Lipopolysaccharide ( LPS ) 10 mg/L inrncreased the proliferation and endothelin release in cultured rnmesangial cells. When mesangial cells pretreated with rn3, 10, and 30 mg/L of salvianolate were incubated for rn4 h with LPS, salvianolate exhibited a concentration dernpendent inhibitory effect on proliferation and endothelin rnlevels in the mesangial cells induced by LPS. Furtherrnmore, the increased basal levels of mesangial cells prolifrneration and the endothelin release were also effectively rninhibited by salvianolate 30 mg/L at 4, 8, and 12 h.rnBesides, no cytotoxicity of salvianolate was observed.rnCONCLUSION: These results indicate that salvianolate rncan inhibit mesangial cells proliferation, which may be rnrelated to the decrease of endothelin release.%目的:研究丹参多酚酸盐对体外培养的大鼠系膜细胞增殖及内皮素释放的影响.方法:细胞增殖的测定用[3H]-TdR掺入法;用放射免疫法测定细胞上清液中内皮素浓度的变化;细胞毒性的测定用MTT及LDH法.结果:LPS 10 mg/L能诱导系膜细胞增殖及内皮素释放的增多.丹参多酚酸盐3、10及30mg/L与系膜细胞孵育4 h,丹参多酚酸盐能浓度依赖性抑制由LPS诱导引起的系膜细胞增殖及内皮素释放的增多.丹参多酚酸盐30 mg/L也能显著抑制系膜细胞4、8和12 h时的增殖及内皮素的释放.丹参多酚酸盐对系膜细胞没有细胞毒作用.结论:丹参多酚酸盐能抑制大鼠系膜细胞的增殖,其机制可能与抑制内皮素的释放有关.
