大豆球蛋白提取条件优化及7S和11 S亚基的鉴定

         

摘要

Soybean glycinin,which is one of the major compositions in soybean meal,greatly influences the quality of soybean meal,and the most important components of glycinin are 7s-conglycinin globulins and 11S globulins.The objective of the study was to decide the optimal conditions for extraction of soybean protein and to analyze 7s-conglycinin globulins and 11S globulins.The results of SDS-PAGE showed that 0.05 M Tris-HCl ( pH9.0,55 ℃) was the optimal condition for the extraction of soybean glycinin with the sample-solution ratio 1 ∶10, and 7s-conglycinin globulins were composed of three subunits with molecular weight of 60466Da, 55029Da and 49043Da,respectively and 11S globulins were also composed of three subunits with molecular weight of 44634Da,39779Da and 17952Da,respectively.%大豆球蛋白是豆粕饲料的主要营养成分之一,对豆粕饲料的营养品质有着重要影响,其中7S伴大豆球蛋白及11S球蛋白是大豆球蛋白最重要的成分。研究分析了在不同提取液,不同的pH,不同的温度,不同浓度的Tris-HCl提取液及不同料液比对豆粕大豆球蛋白提取的影响,并且对7S伴大豆球蛋白和11S球蛋白进行了分析。最终SDS-PAGE结果显示55℃、0.05 mol/L、pH9.0的Tris-HCl溶液在料液比为1∶10时能够提取较多的球蛋白,并且7S伴大豆球蛋白由3个亚基组成,其相对分子量分别为60466 Da,55029 Da和49043 Da,11S球蛋白也由3个亚基组成,其相对分子量分别为44634 Da,39779 Da和17952 Da。
