江西柿属种质资源遗传多样性的IRAP标记分析

The genetic diversities of 49 germplasm resources in Diospyros in Jiangxi were analyzed by using IRAP (Inter-retrotransposon Amplified Polymorphism) molecular marker.Ten primers which had clear bands and polymorphism in PCR amplification were screened out from 26 IRAP primers,and the PCR-amplified products presented clear fingerprint by polyacrylamide gel electmphoresis (PAGE).The 418 clear bands (100％ diversity) were transformed into 0/1 matrix,and the clustering analysis showed that the genetic similarity coefficients among the tested 49 germplasm resources varied from 0.69 to 0.92,and these germplasm resources belonged to 3 species (Diospyros kaki Thunb.,Diospyros oleifera Cheng,and Diospyros kaki var.silvestis Makino),which revealed abundant genetic diversity.In addition,the species taxonomic relationships among several Diospyros spp.germplasm resources were clarified.%利用逆转座子间扩增多态性(Inter-retrotransposon Amplified Polymorphism,IRAP)分子标记对收集的49份江西柿属种质资源进行了遗传多样性分析.从26个IRAP引物中筛选出扩增条带清晰且多态性丰富的10个引物,PCR扩增产物经聚丙烯酰胺凝胶电泳(PAGE)得到清晰的指纹图谱.将418条清晰条带(多态性100％)转换为0/1矩阵,聚类分析结果表明:供试的江西柿属种质资源的遗传相似系数在0.69～ 0.92之间,分属柿、油柿、野柿三个种,遗传多样性丰富;明晰了几个分类关系模糊的柿种质资源的种属关系.
